Fragment-to-Lead Medicinal Chemistry Publications in 2020

Iwan J. P. de Esch et al.

Summary: The review summarizes successful fragment-to-lead studies published in 2020 and discusses trends and best practices in fragment libraries, target proteins, screening technologies, hit-optimization strategies, and properties of hit fragments and leads. Trends and innovations identified in the review promise to further increase the success of FBDD by developing novel screening fragments, improving screening technologies, using computer-aided design and virtual screening, and combining FBDD with other drug-discovery technologies.

Escape from planarity in fragment-based drug discovery: A synthetic strategy analysis of synthetic 3D fragment libraries

Hanna F. Klein et al.

Summary: This review discusses the synthetic strategies for preparing 3D fragments based on 25 papers published from 2011 to mid-May 2020, emphasizing diversity-oriented synthesis, scaffold synthesis and diversification, as well as computational design and synthesis. The conclusion suggests that a workflow combining computational design and other strategies, along with considerations of fragment properties and 3D shape, could promote the wider use of 3D fragments in fragment libraries and facilitate fragment-to-lead optimization.

Simple and modestly scalable synthesis of iso-Cyrene from levoglucosenone and its comparison to the bio-derived and polar aprotic solvent Cyrene®

Xin Liu et al.

Summary: The bio-derived platform molecule levoglucosenone has been converted to its pseudo-enantiomer and then reduced to iso-Cyrene. A less effective synthesis of iso-Cyrene from D-glucose is also discussed. The physicochemical and toxicological properties of iso-Cyrene are compared to those of commercially available Cyrene, revealing significant differences between the two compounds.

Fragment-based drug discovery-the importance of high-quality molecule libraries

Marta Bon et al.

Summary: Fragment-based drug discovery (FBDD) is a complementary approach to high-throughput screening (HTS) that involves screening smaller and less complex molecules. This method has shown promise in optimizing hard-to-drug targets, and recent developments have highlighted its utility against previously undruggable targets.

C-H functionalisation tolerant to polar groups could transform fragment-based drug discovery (FBDD)

Gianni Chessari et al.

Summary: This study analyzed 131 fragment-to-lead examples targeting various protein families between 2015-2019, identifying the most common polar functional groups involved in fragment-protein binding and highlighting the key role of growth from carbocentric vectors. The results suggest that robust C-H functionalisation methods that tolerate the polar functionality on fragments could be transformative for fragment-based drug discovery.
