Sono-ReCORMs for synergetic sonodynamic-gas therapy of hypoxic tumor

Yue Li et al.

Summary: Carbon monoxide (CO) gas therapy, a new anti-tumor technique, has attracted attention in cancer treatment due to its cytotoxicity. This technique shows promising therapeutic effect for hypoxic tumors, where traditional photodynamic therapy is ineffective. To overcome the limitations of photodynamic therapy, a synergetic sonodynamic-CO gas releasing strategy is proposed. The rhenium(I) tricarbonyl complex with an electron-donating ligand exhibits stronger luminescence and ROS generation, as well as CO release triggered by ultrasound, making it a powerful tool for the treatment of hypoxic tumors.

A tricarbonyl rhenium(I) complex decorated with boron dipyrromethene for endoplasmic reticulum-targeted photodynamic therapy

Wen-Wen Feng et al.

Summary: Organelle-targeted photodynamic therapy (PDT) using a tricarbonyl Re(I) complex (Re-BDP) was shown to effectively kill cancer cells by generating toxic reactive oxygen species (ROS) in the endoplasmic reticulum (ER). Re-BDP exhibited strong absorbance and high singlet oxygen quantum yield, and preferentially accumulated in the ER. Upon visible light irradiation, Re-BDP showed high phototoxicity and induced caspase-dependent apoptosis accompanied by mitochondrial dysfunction. This study highlights the potential of developing efficient organelle-targeted phototherapeutic agents by conjugating metal complexes and organic chromophores.

Protoporphyrin-sensitized degradable bismuth nanoformulations for enhanced sonodynamic oncotherapy

Kang Song et al.

Summary: In order to enhance the efficacy of low-frequency sonodynamic therapy, we developed a protoporphyrin-sensitized bismuth-based nano-semiconductor as a sonosensitizer. This material can reduce the scavenging capacity of reactive oxygen species and increase their production, leading to improved therapeutic efficacy against tumors. Additionally, it can deplete glutathione and break the redox balance of the tumor microenvironment, further enhancing the effect of sonodynamic therapy.
