GSDME Is Related to Prognosis and Response to Chemotherapy in Oral Cancer

S. Wang et al.

Summary: Gasdermin E (GSDME) is highly expressed in oral squamous cell carcinoma (OSCC) and is associated with better prognosis. Chemotherapy drugs can induce GSDME-mediated pyroptosis, and the loss of GSDME function attenuates the anti-tumor effect of cisplatin.

CXCR4-targeted nanotoxins induce GSDME-dependent pyroptosis in head and neck squamous cell carcinoma

Elisa Rioja-Blanco et al.

Summary: Novel therapeutic strategies are urgently needed for HNSCC patients with therapy resistance. T22-PE24-H6 and T22-DITOX-H6 nanotoxins show promising potential for the treatment of HNSCC by specifically targeting CXCR4-overexpressing cells and triggering caspase-3/GSDME-dependent pyroptosis.

Biomaterial-mediated modulation of oral microbiota synergizes with PD-1 blockade in mice with oral squamous cell carcinoma

Di-Wei Zheng et al.

Summary: It has been demonstrated that modulating the microbiota in tumors can enhance antitumor immune responses and improve cancer patient survival rates. The use of hydrogel-embedded silver nanoparticles and specific exogenous bacteria shows potential in boosting the effectiveness of cancer therapies by influencing the host's immune system through host-microbiota interactions.

Overexpression of RRM2 is related to poor prognosis in oral squamous cell carcinoma

Shuo Wang et al.

Summary: The overexpression of RRM2 in oral squamous cell carcinoma is associated with disease progression, recurrence, and poor prognosis. RRM2 expression promotes cancer cell proliferation and migration while inhibiting apoptosis. This suggests that RRM2 could be a potential target for diagnosis, prognosis, and therapy of OSCC.

Synergism of TNF-α and IFN-γ Triggers Inflammatory Cell Death, Tissue Damage, and Mortality in SARS-CoV-2 Infection and Cytokine Shock Syndromes

Rajendra Karki et al.

Summary: The combination of TNF-alpha and IFN-gamma induces inflammatory cell death characterized by PANoptosis, which can be inhibited to protect mice from the pathological effects and death caused by COVID-19. Blocking the cytokine-mediated inflammatory cell death signaling pathway may benefit patients with COVID-19 or other infectious and autoinflammatory diseases by limiting tissue damage/inflammation.

A pyroptosis-related lncRNA signature predicts prognosis and immune microenvironment in head and neck squamous cell carcinoma

Wenying Zhu et al.

Summary: This study identified a potential clinical value of pyroptosis-related long non-coding RNAs in head and neck squamous cell carcinoma. The lncRNA signature consisting of 14 pyroptosis-related long non-coding RNAs could serve as a prognostic marker for HNSCC, suggesting pyroptosis as a promising therapeutic target in this type of cancer.

Fueling the Fire: Inflammatory Forms of Cell Death and Implications for Cancer Immunotherapy

Sheera R. Rosenbaum et al.

Summary: Using immune checkpoint inhibitors (ICI) has significantly improved overall survival for certain cancer patients, but many tumors do not respond due to a lack of tumor-infiltrating lymphocytes (TIL). Converting immune cold tumors to hot tumors is a key obstacle in cancer treatment. Promoting tumor-localized necroptosis and pyroptosis may enhance responses to ICI.

Microenvironment-Responsive Prodrug-Induced Pyroptosis Boosts Cancer Immunotherapy

Yao Xiao et al.

Summary: The absence of tumor antigens poses a challenge in ICB therapy, but inducing tumor-specific pyroptosis while reducing injury in normal tissues is urgently needed. A ROS/GSH dual-responsive nano-prodrug called MCPP with high PTX and P18 loading is designed to selectively induce pyroptosis in tumor cells. Chemo-photodynamic therapy and controlled-release PTX synergistically induce pyroptosis, releasing DAMPs and enhancing ICB efficiency, suggesting a smart strategy to trigger pyroptosis and augment ICB efficiency.

Prognostic role of DFNA5 in head and neck squamous cell carcinoma revealed by systematic expression analysis

Summary: The study found that DFNA5 expression is prognostic for patients with HNSCC, with higher levels associated with worse survival. DFNA5 may function in HNSCC by regulating lymphocyte infiltration.

Human TBK1 deficiency leads to autoinflammation driven by TNF-induced cell death

Tumor necrosis factor-alpha and interferon-gamma induce inflammasome-mediated corneal endothelial cell death

Angela Gomez et al.

Summary: Chronic corneal endothelial cell loss, induced by pro-inflammatory cytokines like TNF-alpha and IFN-gamma, is associated with inflammasome activation and subsequent pyroptosis. Inhibition of inflammasome presents a promising therapeutic target for the treatment of chronic CEC loss.
