Interaction analyses of hTAAR1 and mTAAR1 with antagonist EPPTB

Summary: Trace amine-associated receptor 1 (TAAR1) plays a critical role in regulating monoaminergic activity. EPPTB is a potent antagonist of mouse TAAR1 but weakly antagonizes human TAAR1. This study used computational methods to explore the interaction modes between EPPTB and TAAR1. The binding affinity of EPPTB to human TAAR1 is weaker than to mouse TAAR1, possibly due to differences in the binding site residues. Another antagonist, Compound 3, shows slightly stronger binding affinity to human TAAR1 and binds to a similar pocket.

Influenza Polymerase Inhibitors: Mechanisms of Action and Resistance

Summary: The RNA-dependent RNA polymerase of influenza virus is highly conserved among different types of influenza viruses and is considered a key target for antiviral agents. Several polymerase inhibitors targeting different subunits have been developed, including PB1 inhibitor favipiravir, PB2 inhibitor pimodivir, and PA inhibitor baloxavir marboxil, which have shown antiviral activity and are in late-phase clinical trials for influenza treatment.
