Dissecting the molecular basis of human interneuron migration in forebrain assembloids from Timothy syndrome

Fikri Birey et al.

Summary: In this study, it was found that the role of L-type calcium channel in neurodevelopmental diseases is complex, as it can regulate the saltation length and frequency of interneuron migration. Inhibition of GABA-A receptors can restore the defect in migration frequency, while modulation of actomyosin and myosin light chain phosphorylation can improve the defect in saltation length.

Maturation and circuit integration of transplanted human cortical organoids

Omer Revah et al.

Summary: This study demonstrates the potential of transplanting self-organizing neural organoids derived from human stem cells into the somatosensory cortex of rats to develop mature cell types and integrate into sensory and motivation-related circuits. The transplanted cortical neurons exhibit more complex properties than their in vitro counterparts and can drive reward-seeking behavior when optogenetically activated. This approach has the potential to detect circuit-level phenotypes in patient-derived cells that cannot be uncovered using other methods.

A single-cell transcriptome atlas of glial diversity in the human hippocampus across the postnatal lifespan

Yijing Su et al.

Summary: This study used single-nucleus RNA sequencing to analyze the transcriptome atlas of glia in the human hippocampus. The results revealed distinct molecular signatures of glial subpopulations and their associations with physiological functions, age-dependent changes, and relevance to diseases such as Alzheimer's disease. Furthermore, the study characterized the spatiotemporal heterogeneity of certain astrocyte subpopulations and identified dysregulated genes and pathological processes in specific glial subpopulations in Alzheimer's disease. The findings significantly contribute to our understanding of human glial diversity and the dysregulation in Alzheimer's disease, as well as provide a reference atlas for stem cell-based glial differentiation.
