Stress-induced depressive-like behavior in male rats is associated with microglial activation and inflammation dysregulation in the hippocampus in adulthood

Joao Paulo Bras et al.

Summary: This study suggests that male rats with high-CORT stress responsiveness exhibit increased anhedonia and passive coping responses in adulthood following peripubertal stress exposure. Additionally, exposure to peripubertal stress leads to increased hippocampal TNF-alpha expression, which correlates with passive coping responses. Furthermore, peripubertal stress has long-term effects on hippocampal microglia, particularly in high-CORT stress-responsive rats.

Dysregulation of mitochondrial dynamics, mitophagy and apoptosis in major depressive disorder: Does inflammation play a role?

Giselli Scaini et al.

Summary: Recent studies have shown that mitochondrial dysfunction and dysregulated neuroinflammatory pathways play a role in major depressive disorder (MDD). This study found differences in markers of mitochondrial dynamics, mitophagy, autophagy, and apoptosis in MDD patients compared to healthy controls, with higher levels of specific proteins associated with mitochondrial dysfunction in MDD patients. Furthermore, inflammation and protein levels related to mitochondrial dynamics and mitophagy were found to influence the severity of depressive symptoms in MDD patients.

Sustained TNF signaling is required for the synaptic and anxiety-like behavioral response to acute stress

Gina M. Kemp et al.

Summary: Acute stress induces synaptic plasticity and behavioral changes in the brain. The study reveals that TNF is a crucial mediator in the stress response of mice, required for stress-induced synaptic potentiation and increased anxiety-like behavior. The activation of microglia leads to the long-term release of TNF, while sustained TNF signaling in the ventral hippocampus is necessary for maintaining the stress-induced changes.

BNIP3L/NIX degradation leads to mitophagy deficiency in ischemic brains

Xiaoli Wu et al.

Summary: Mitophagy is essential for promoting neuronal survival in cerebral ischemia, and the deficiency of mitophagy receptor BNIP3L/NIX can lead to increased brain injury in ischemic conditions. The degradation of BNIP3L by proteasomes in ischemic brains contributes to mitophagy deficiency, and pharmacological intervention targeting this process, such as using the proteasome inhibitor carfilzomib, can rescue mitophagy and protect against ischemic brain injury.

Transcription Factor TWIST1 Integrates Dendritic Remodeling and Chronic Stress to Promote Depressive-like Behaviors

Jin-Gang He et al.

Summary: The study found that the expression of TWIST1 in the brain is positively correlated with depressive behaviors, chronic stress can elevate TWIST1 expression in the medial prefrontal cortex of mice which can be reversed by fluoxetine treatment. Overexpression of TWIST1 increases susceptibility to stress, while knockdown of TWIST1 prevents defective dendrite morphogenesis in mice and alleviates depressive-like behaviors. The prodepressant property of TWIST1 is mediated by the repression of miR-214-PPAR-delta signaling and mitochondrial function, which can be mimicked by genetic and pharmacological inhibition of PPAR-delta.

Chronic Stress Induces Sex-Specific Functional and Morphological Alterations in Corticoaccumbal and Corticotegmental Pathways

Summary: This study found that chronic stress induces depressive-like behaviors in both male and female animals. NAc- and VTA-projecting mPFC neurons exhibited sex-specific changes, with more severe functional and morphological alterations in females in the NAc pathway and more drastic reduction in dendritic complexity in males in the VTA pathway. Chemogenetic overactivation of the corticoaccumbal pathway triggered anxiety and behavioral despair in both sexes, while its inhibition only rescued the phenotype in females.

TNF leads to mtDNA release and cGAS/STING-dependent interferon responses that support inflammatory arthritis

Joschka Willemsen et al.

Summary: The study elucidates a molecular mechanism linking TNF to type-I interferon signaling, demonstrating that TNF triggers a cGAS-STING-dependent interferon response and inhibits PINK1-mediated mitophagy. This leads to altered mitochondrial function, increased cytosolic mtDNA levels, and a transcriptional response that mimics macrophages from rheumatoid arthritis patients. Additionally, in an inflammatory arthritis mouse model, cGAS deficiency blocked interferon responses and reduced inflammatory cell infiltration and joint swelling. Therapeutic targeting of cGAS/STING in TNF-driven diseases may offer potential benefits.

The role of mitophagy in the regulation of mitochondrial energetic status in neurons

Sinsuk Han et al.

Summary: Mitochondria are crucial for supplying energy to neurons, and dysfunction in these organelles is associated with early features of Alzheimer's disease (AD). In AD, metabolic and mitochondrial disruptions occur before clinical symptoms, and mitophagy, a process of clearing damaged mitochondria, plays a role in maintaining energy metabolism and synaptic function. However, in AD mouse models, early stimulation of mitochondrial activity can worsen synaptic loss due to impaired mitophagy. Enhancing lysosomal function restores metabolic function and protects against synaptic damage in AD. This research suggests that targeting mitophagy and lysosomal activity may offer potential therapeutic strategies for AD.
