SunUp and Sunset genomes revealed impact of particle bombardment mediated transformation and domestication history in papaya

Jingjing Yue et al.

Summary: This study investigated the genome structure of transgenic papaya and the impact of particle bombardment on the genome. The results revealed that the genomes of transgenic papaya SunUp and its progenitor Sunset consist of nine chromosomes. A segment containing three transgenic insertions was identified in one of the chromosomes of SunUp. Additionally, the study discovered the genome structure of the hermaphrodite-specific region and its X counterpart. Resequencing analysis demonstrated the geographic origin and breeding history of papaya.

Impacts of RNA Mobility Signals on Virus Induced Somatic and Germline Gene Editing

Bliss M. M. Beernink et al.

Summary: The impact of RNA mobility signals on virus-induced somatic and germline gene editing in plants is unclear. While adding certain signals can enhance somatic mutations, they are not sufficient to induce germline mutations. Furthermore, other viral guide RNA delivery systems are also capable of inducing somatic and germline mutations, suggesting the importance of virus type and sgRNA specificity in the editing process.

Summary: The development of a PVX-based vector allows for the easy and efficient expression of multiple sgRNAs for multiplex CRISPR-Cas genome editing in adult Solanaceae plants, achieving nearly 80% indels in targeted genes within a few days. The vector also enables the production of virus-free edited progeny with high rates of heritable biallelic mutations, making it a valuable tool for functional gene analysis and precision breeding in diverse plant species, particularly Solanaceae crops.

Heritable gene editing using FT mobile guide RNAs and DNA viruses

Jianfeng Lei et al.

Summary: The virus-induced genome editing (VIGE) system has great application potential, but it cannot transfer edited genes to the next generation due to the inability of viruses to enter shoot apical meristem (SAM) in plants. A novel CLCrV-mediated VIGE system using the FT strategy enables heritable mutant offspring without tissue culture in A. thaliana.
