Design, Synthesis, and biological evaluation of HDAC6 inhibitors based on Cap modification strategy

Summary: The abnormal biological functions of HDAC6 are closely related to the occurrence and development of various tumors, making it a promising therapeutic target for cancer treatment. In this study, 20 compounds were designed and synthesized to assess their biological activities against tumor cells. Compounds 9m, 9q, and 12c showed promising inhibitory activities, especially against HDAC6. Furthermore, the binding modes of compounds 9q and 12c in HDAC1&6 were investigated computationally, providing valuable insights into the discovery of novel HDAC6 inhibitors.
