Fructose impairs fat oxidation: Implications for the mechanism of western diet-induced NAFLD

Mustafa Kutlu Inci et al.

Summary: Increased fructose intake, especially in combination with a high-fat diet, can lead to obesity and metabolic complications. Fructose impairs the oxidation of dietary fat by affecting the mitochondrial beta-oxidation pathway and inhibiting the production of acylcarnitine. This explains the synergistic effects of fructose and fat on the development of conditions like non-alcoholic fatty liver disease.

NRF2 and FXR dual signaling pathways cooperatively regulate the effects of oleanolic acid on cholestatic liver injury

Jianming Liu et al.

Summary: This study found that the effect of oleanolic acid (OA) on cholestatic liver injury is closely related to the simultaneous activation of NRF2 and FXR dual signaling pathways, in which the NRF2 signaling pathway plays a more important role. The dual signaling pathways cooperatively regulate bile acid metabolic homeostasis through the interaction mechanism with β-catenin/P300.

TGF-β1 promotes SCD1 expression via the PI3K-Akt-mTOR-SREBP1 signaling pathway in lung fibroblasts

Zili Zhou et al.

Summary: This study found that lung fibroblast activation is associated with airway remodeling during asthma progression. In a mouse asthma model, the inhibition of SCD1 significantly improved airway remodeling and lung fibroblast activation. The study also found that the PI3K-Akt-mTOR-SREBP1 pathway is involved in the regulation of SCD1 expression and lung fibroblast activation. Therefore, the inhibition of SCD1 may be a therapeutic strategy for airway remodeling in asthma.

Ursolic Acid Analogs as Potential Therapeutics for Cancer

Siva S. Panda et al.

Summary: Ursolic acid (UA) is a pentacyclic triterpene with diverse pharmacological properties, including anti-inflammatory, antimicrobial, antiviral, antioxidant, and anti-proliferative activities. It holds promise as a cancer preventive and therapeutic agent due to its relatively non-toxic properties against normal cells but its activities against cancer cells. However, its low bioavailability limits its clinical applications. Researchers have been designing and developing synthetic analogs of UA to overcome this limitation and utilize its importance.

Lipid desaturation regulates the balance between self-renewal and differentiation in mouse blastocyst-derived stem cells

Chanchal Thomas Mannully et al.

Summary: Stem cell self-renewal and differentiation are regulated by metabolism, specifically lipid desaturation. Inhibiting specific metabolic enzymes can maintain stem cell characteristics and reduce cell apoptosis. Monounsaturated fatty acids are beneficial for stem cells but detrimental to trophoblast cells.

Chiara De Stefani et al.

Summary: The study formulated oleanolic acid (OA) into two different microemulsions (ME-1 and ME-2) to enhance its solubility and intestinal permeability, leading to a significant increase in bioavailability. Both MEs showed enhanced intestinal permeability compared to free OA according to PAMPA assay results. The study suggests that MEs could be a promising formulation to improve the oral bioavailability of OA with potential clinical applications.

Non-alcoholic fatty liver disease

Elizabeth E. Powell et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is a common global health issue with significant associations with metabolic syndrome and type 2 diabetes, leading to serious complications like cirrhosis and hepatocellular carcinoma. While cardiovascular disease and extrahepatic malignancy are the main causes of death in NAFLD patients, advanced liver fibrosis is a key prognostic marker for liver-related outcomes and overall mortality.
