Ceiling Effects and Differential Measurement Precision Across Calibrated Cognitive Scores in the Framingham Study

Phoebe Scollard et al.

Summary: The objective of this study was to calibrate cognitive assessment data from the Framingham Heart Study (FHS) across different waves, addressing design considerations, ceiling effects, and measurement precision. Cognitive tests were assigned to specific domains and calibrated using bifactor confirmatory factor analysis models. The results indicated that memory domain scores were the most precisely estimated.

Progranulin mutations in clinical and neuropathological Alzheimer's disease

Badri N. Vardarajan et al.

Summary: This study found that pathogenic GRN mutations are common in FTLD and AD, often with TDP-43 pathology. Pathogenic GRN carriers have significantly higher PHFtau tangle density associated with AD. The rs5848 mutation is associated with increased frequency of hippocampal sclerosis and TDP-43 deposits in AD patients.

Progranulin as a therapeutic target in neurodegenerative diseases

Herve Rhinn et al.

Summary: Progranulin plays a key role in the development and maintenance of neurons and microglia in the brain. Mutations in the GRN gene can cause neurodegenerative diseases, while mutations that reduce PGRN levels increase the risk of other neurological disorders. Elevating and restoring PGRN levels is an attractive therapeutic strategy for neurodegenerative diseases.

Sex differences in the genetic architecture of cognitive resilience to Alzheimer's disease

Jaclyn M. Eissman et al.

Summary: Approximately 30% of elderly adults with Alzheimer's disease neuropathology show no cognitive decline. This study investigated the genetic factors contributing to resilience to cognitive consequences of Alzheimer's disease neuropathology in a sex-specific manner. The results identified a female-specific resilience locus and highlighted numerous sex-specific molecular pathways related to resilience. These findings suggest the importance of personalized treatment based on sex and genetic factors.

Aβ42 oligomers trigger synaptic loss through CAMKK2-AMPK-dependent effectors coordinating mitochondrial fission and mitophagy

Annie Lee et al.

Summary: This study reveals that soluble Amyloid-beta 1-42 oligomers (A beta 42o) trigger the loss of excitatory synapses in early Alzheimer's disease, prior to the formation of insoluble amyloid plaques. The researchers also found that A beta 42o leads to structural remodeling of mitochondria in specific regions and mediates synaptic loss through certain signaling pathways.

The genetic architecture of human complex phenotypes is modulated by linkage disequilibrium and heterozygosity

Dominic Holland et al.

Summary: The study introduces an extended Gaussian mixture model for explaining the distribution of causal effects for human complex phenotypes. It finds that causal effects are distributed with dependence on total LD and H, with SNPs having lower LD and H more likely to have larger effects, consistent with models of negative pressure from natural selection.

Multi-trait association studies discover pleiotropic loci between Alzheimer's disease and cardiometabolic traits

William P. Bone et al.

Summary: Genetic risk factors shared between Alzheimer's disease (AD) and cardiometabolic traits were identified in this study, revealing a complex genetic relationship between them. The results suggest that blood pressure and immune response play a role in the pleiotropy between these traits, potentially providing clues for discovering new therapeutic targets.

A genome-wide association study with 1,126,563 individuals identifies new risk loci for Alzheimer's disease

Douglas P. Wightman et al.

Summary: This study identified seven new genetic loci associated with Alzheimer's disease through a genome-wide association study, implicating microglia and immune cells in late-onset disease. The research highlights the relevance of microglia, immune cells, and protein catabolism to late-onset Alzheimer's disease, while prioritizing previously unidentified genes for further investigation. The results are expected to contribute to larger meta-analyses to uncover additional genetic variants affecting Alzheimer's pathology.

Summary: The study utilized a new statistical method, PLACO, to identify genetic overlap between cleft lip with or without cleft palate (CL/P) and cleft palate alone (CP). The research discovered new genetic loci influencing risk for both CL/P and CP, with locus-specific effects varying by racial/ethnic group. Furthermore, the study confirmed shared genetic etiology of subtypes underlying CL/P, while also revealing new loci of genetic overlap exhibiting etiologic differences.

Differential patterns of gray matter volumes and associated gene expression profiles in cognitively-defined Alzheimer's disease subgroups

Colin Groot et al.

Summary: In this study, differences in regional gray matter volumes and gene expression profiles were compared between cognitively-defined subgroups of individuals with AD, revealing neurobiological distinctions associated with each subgroup's cognitive profile. These findings suggest that distinct biological pathways may contribute to the emergence of cognitive heterogeneity in AD.
