The Cerebellum in Niemann-Pick C1 Disease: Mouse Versus Man

Maria Teresa Fiorenza et al.

Summary: Selective neuronal vulnerability is a common feature in degenerative disorders. This article focuses on the vulnerability of the cerebellum in Niemann-Pick C (NPC), discussing the impact of altered intracellular cholesterol trafficking on the development and functional maturation of cerebellar cells. The article also addresses the progression of cerebellar defects and the persistence of these defects in mouse models despite genetic manipulations.

Disruptive lysosomal-metabolic signaling and neurodevelopmental deficits that precede Purkinje cell loss in a mouse model of Niemann-Pick Type-C disease

Sarah Kim et al.

Summary: Purkinje cell (PC) loss occurs early in Niemann-Pick Type C (NPC) disease, caused by mutations in the Npc1 or Npc2 genes. This study found that NPC1 deficiency affects the postnatal development of PC dendrites and synapses, leading to metabolic imbalance. The disruption of lysosomal-metabolic signaling in PCs causes dendritic and synaptic developmental deficits that precede and promote their early degeneration in NPC.

Developmental timing-dependent organization of synaptic connections between mossy fibers and granule cells in the cerebellum

Taegon Kim et al.

Summary: Recent findings support the hypothesis that synapses between mossy fibers (MFs) and cerebellar granule cells (GCs) are organized based on the origins of MFs and locations of GC axons, parallel fibers (PFs). The mechanisms of this organization are still unknown. Using a new labeling technique, researchers confirmed that synaptic connections of GCs with specific MFs were orderly organized according to their PF locations. They also observed a bias in overall MF-GC synaptic connectivity, where GC dendrites with nearby PFs tended to connect with the same MF terminals. The study suggests that this organization is likely the result of synaptic formation between developmentally matched partners.

Pleiotropic effects of BDNF on the cerebellum and hippocampus: Implications for neurodevelopmental disorders

Serena Camuso et al.

Summary: Brain-derived neurotrophic factor (BDNF) is a highly studied neurotrophin in the mammalian brain, crucial for the development of neurons and synaptic plasticity. It is expressed at higher levels in brain areas such as cerebellum and hippocampus. Dysregulation of BDNF expression is associated with neurodevelopmental disorders. Understanding the role of BDNF is important for researching and treating these disorders.

The appearance of phagocytic microglia in the postnatal brain of Niemann Pick type C mice is developmentally regulated and underscores shortfalls in fine odor discrimination

Alessandro Rava et al.

Summary: The loss of NPC1 or NPC2 function leads to dysregulation of lipid metabolism, affecting brain development and causing pathological symptoms. Microglia activation is a key event in NPC1 disease progression, but the mechanisms of how microglia respond to neurodevelopmental changes are still not fully understood.

BDNF haploinsufficiency induces behavioral endophenotypes of schizophrenia in male mice that are rescued by enriched environment

Mahmoud Harb et al.

Summary: BDNF is important for brain function, and low levels are associated with schizophrenia. A study on BDNF+/- mice showed deficits in attention, startle response, and sensorimotor gating, which were prevented by an enriched environment. These findings suggest a relationship between decreased BDNF levels and behavioral endophenotypes of schizophrenia.

Neurotrophins as Key Regulators of Cell Metabolism: Implications for Cholesterol Homeostasis

Mayra Colardo et al.

Summary: Neurotrophins are a family of growth factors initially thought to primarily mediate nervous system development and neuronal survival, but increasing evidence suggests they also play crucial roles in regulating metabolic processes such as redox balance, glucose homeostasis, and cholesterol metabolism. This review examines the signaling pathways of neurotrophins and their impact on metabolism, particularly focusing on their effects on cholesterol metabolism.
