The Role of RNA Secondary Structure in Regulation of Gene Expression in Bacteria

PlzA is a bifunctional c-di-GMP biosensor that promotes tick and mammalian host-adaptation of Borrelia burgdorferi

Summary: This study investigated the roles of c-di-GMP and its only known effector protein, PlzA, in Borrelia burgdorferi during the arthropod and mammalian phases of the enzootic cycle. The protective function of PlzA in ticks was confirmed to be c-di-GMP-dependent, while a c-di-GMP-independent function in mammals was also identified. Loss of PlzA did not affect expression of RpoS or RpoS-dependent genes, and enhanced host-adaptation in the absence of PlzA contributed to restored infectivity. Structural analysis revealed conformational changes in PlzA upon c-di-GMP binding that underlie its dual functionality.
