Experimental validation of taxon-specific mini-barcode primers for metabarcoding of zooplankton

Ashrenee Govender et al.

Summary: Metabarcoding is a fast-developing field used to determine the species composition and diversity of marine zooplankton communities, but standardization of methods is still incomplete. The choice of genetic markers and primers is crucial for influencing species detection rates and accuracy. Taxon-specific mini-barcode primers have been developed to enhance species detection rates and accuracy in this study.

Population and molecular responses to warming in Netzelia tuberspinifera - An endemic and sensitive protist from East Asia

Wenping Wang et al.

Summary: Research has shown that rising temperatures significantly affect the population changes of the sensitive species N. tuberspinifera, with transcriptomic analysis revealing different molecular responses at various temperatures. The study also found that N. tuberspinifera is unable to survive when temperatures exceed 40 degrees Celsius.

Multiple convergences in the evolutionary history of the testate amoeba family Arcellidae (Amoebozoa: Arcellinida: Sphaerothecina): when the ecology rules the morphology

Ruben Gonzalez-Miguens et al.

Summary: Arcellidae, a diverse family of amoebae, is found worldwide in various environments and serves as an indicator species for environmental quality. However, their systematics are based on outdated research, with morphology and ecological data suggesting that shell shape may be influenced more by environmental factors rather than phylogenetic relationships. The study also reveals convergent evolution in species with similar ecology, and proposes the reevaluation and description of several new species and a new genus.

Mitochondrial Cytochrome Oxidase Subunit 1: A Promising Molecular Marker for Species Identification in Foraminifera

Elsa B. Girard et al.

Summary: Traditional morphological methods for species identification are time-consuming. To analyze more samples efficiently, molecular tools have been extended to species identification. A new mitochondrial marker has been discovered, showing higher potential for species identification compared to existing markers. This marker has a higher success rate in amplification and better performance in genetic variability. Moreover, it helps recognize difficult-to-morphologically-identify specimens.

Discrepancies between prokaryotes and eukaryotes need to be considered in soil DNA-based studies

Enrique Lara et al.

Summary: Metabarcoding approaches have greatly contributed to our understanding of soil biodiversity, but direct transfer of methods between prokaryotic and eukaryotic soil metabarcoding is currently not possible due to fundamental differences. Future optimization of eukaryotic metabarcoding includes improved qualitative and quantitative characterization of eukaryotes.

Cyphoderia ampulla (Cyphoderiidae: Rhizaria), a tale of freshwater sailors: The causes and consequences of ecological transitions through the salinity barrier in a family of benthic protists

Ruben Gonzalez-Miguens et al.

Summary: The study shows that crossing the salinity barrier allows organisms to access new ecosystems and experience significant shifts in diversification rates while adapting to changes in selective pressures. Analysis suggests that freshwater environments have triggered four independent transitions since the mid-Miocene, with ancestral euryhaline species adapting to fluctuating salinity levels.

Deconstructing Difflugia: The tangled evolution of lobose testate amoebae shells (Amoebozoa: Arcellinida) illustrates the importance of convergent evolution in protist phylogeny

Ruben Gonzalez-Miguens et al.

Summary: In this study, the authors used Arcellinida testate amoebae as a model to test the paradigm of morphological stasis in protists. The results showed a lack of conservatism in shell outlines and widespread morphological convergences between far-related taxa. In addition, the authors revised the systematics and established a new infraorder and two new genera.

Metabarcoding of fecal DNA shows dietary diversification in wolves substitutes for ungulates in an island archipelago

Gretchen H. Roffler et al.

Summary: The study found that wolves alter their dietary diversity and foraging patterns in response to the availability of their main prey, ungulates. In the Alexander Archipelago, wolves primarily consume ungulates, but when ungulates become scarce, they increase the number and diversity of prey species consumed instead of focusing on a few specific items.

Diversity of Testate Amoebae as an Indicator of the Conservation Status of Peatlands in Southwest Europe

Rafael Carballeira et al.

Summary: Testate amoebae are important bioindicators of Sphagnum peatland ecosystems, which are threatened wetland types with a delicate biogeochemical balance. A study on chemical parameters and testate amoebae diversity in the western Iberian Peninsula revealed an inverse relationship between diversity and conservation status, influenced by anthropogenic activities and geographical patterns of disturbance.
