Loss of the fragile X syndrome protein FMRP results in misregulation of nonsense-mediated mRNA decay

Tatsuaki Kurosaki et al.

Summary: The loss of fragile X syndrome protein FMRP leads to hyperactivated nonsense-mediated mRNA decay, resulting in defects in neuronal differentiation. The key NMD factor UPF1 directly binds to FMRP, promoting abnormal translation of NMD targets and contributing to molecular abnormalities in FMRP-deficient cells. Inhibition of hyperactivated NMD restores neurodifferentiation markers in FXS patient-derived iPSCs.

Identification of RNA pseudouridine sites using deep learning approaches

Abu Zahid Bin Aziz et al.

Summary: In this paper, a multi-channel convolution neural network using binary encoding was proposed for identifying pseudouridine sites. Tuning hyperparameters with k-fold cross-validation and grid search, promising results were obtained in independent datasets. This method proved to be effective for identifying pseudouridine sites and has been implemented as an easily accessible web server.

Therapeutic promise of engineered nonsense suppressor tRNAs

Summary: Nonsense mutations create defective truncated proteins by changing amino acid codons. While most PTC therapeutics focus on promoting PTC read-through, there is a need for agents that can recode PTCs with the correct amino acids.

Nonsense suppression therapies in human genetic diseases

Patricia Martins-Dias et al.

Summary: Nonsense mutations can lead to dysfunctional proteins, but nonsense suppression therapy has the potential to restore protein function and treat a variety of genetic disorders. However, the efficiency of suppression may be influenced by nonsense-mediated decay, highlighting the importance of using NMD inhibitors or readthrough-compound potentiators to enhance therapeutic effects.
