Coordinate control of basal epithelial cell fate and stem cell maintenance by core EMT transcription factor Zeb1

Yingying Han et al.

Summary: The study reveals the intrinsic requirement of Zeb1 in mammary epithelial ductal side branching and basal cell regenerative capacity. Zeb1 maintains both basal cell fate and stem cell quiescence partly through suppressing Wnt signaling.

Clinical features and prognostic impact of asymptomatic pancreatic cancer

Tetsuya Takikawa et al.

Summary: This study reviewed the data of 577 patients with pancreatic ductal adenocarcinoma (PDAC) and found that the diagnosis of asymptomatic PDAC is associated with early diagnosis and a better prognosis. In asymptomatic PDAC, the tumors are smaller, the disease is in an earlier stage, and the resectability is higher.

Stromal oncostatin M cytokine promotes breast cancer progression by reprogramming the tumor microenvironment

Angela M. Araujo et al.

Summary: This study reveals the pivotal role of the cytokine oncostatin M (OSM) in regulating interactions between immune and nonimmune stromal cells and epithelial cancer cells. It demonstrates that the stromal OSM/OSMR axis reprograms the tumor microenvironment and plays a key role in breast cancer progression.

EMT, cancer stem cells and autophagy; The three main axes of metastasis

Ghader Babaei et al.

Summary: Epithelial-mesenchymal transition (EMT) and Cancer stem-like cells (CSCs) are major factors contributing to cancer cell metastasis. Autophagy has recently gained attention, playing a dual role in cancer cell development depending on cancer type, tissue, and stage. Studies show different signaling pathways are involved in cancer stem cells, autophagy, and EMT. Investigating effective signaling pathways in all three processes could help determine appropriate therapeutic goals.

Exploring the oncostatin M (OSM) feed-forward signaling of glioblastoma via STAT3 in pan-cancer analysis

Miao Chen et al.

Summary: This study conducted a pan-cancer analysis on OSM, showing higher expression of OSM in most tumor tissues compared to normal tissues, with a strong correlation to poor prognosis in several cancers. OSM expression was related to stromal and immune cell infiltration in the tumor microenvironment, as well as immune checkpoint and chemokine co-expression. In GBM, OSM was found to stimulate malignant cells through the Jak-STAT and NF-kappa B pathways, and could facilitate migration and invasion of glioblastoma cells. The study indicates that OSM may serve as a potential immunotherapeutic target for cancer treatment.

Global characterization of macrophage polarization mechanisms and identification of M2-type polarization inhibitors

Lizhi He et al.

Summary: Macrophages undergoing M1 versus M2-type polarization exhibit significant differences in cell metabolism, cellular functions, and signaling pathways, especially in metabolic pathways. Drug screens have identified inhibitors that selectively block M2-type polarization, with MEK signaling being found necessary for M2-type polarization in a proof-of-principle study using comprehensive proteomics and phosphoproteomics datasets.

Characterisation of circulating tumour cell phenotypes identifies a partial-EMT sub-population for clinical stratification of pancreatic cancer

Alexander Semaan et al.

Summary: DEP-FFF was used to isolate CTCs in PDAC patients, revealing four distinct sub-populations including pEMT-CTCs, which were correlated with disease progression, survival, and recurrence. This highlights the importance of immunophenotyping and quantifying specific CTC sub-populations in PDAC.

Dissecting FGF Signalling to Target Cellular Crosstalk in Pancreatic Cancer

Edward P. Carter et al.

Summary: Pancreatic ductal adenocarcinoma (PDAC) has a poor prognosis due to challenges in treatment and late detection, with the FGFR signalling pathway playing a critical role in tumor progression and interaction with the tumor microenvironment. However, it is clear that the FGFR signalling pathway does not act in isolation and interacts with other signaling networks. Therapeutic approaches are being developed to improve prognosis for this terrible disease.

Interactions between cancer cells and immune cells drive transitions to mesenchymal-like states in glioblastoma

Toshiro Hara et al.

Summary: The mesenchymal subtype of glioblastoma is believed to be influenced by both cancer cell-intrinsic alterations and external cellular interactions, with macrophages inducing a transition of glioblastoma cells into mesenchymal-like states. This transition is mediated by macrophage-derived oncostatin M (OSM) interacting with its receptors on glioblastoma cells, leading to the activation of STAT3. This study also reveals that mesenchymal-like glioblastoma states are associated with increased expression of a mesenchymal program in macrophages and enhanced cytotoxicity of T cells, suggesting potential therapeutic implications.

Novel mechanism for OSM-promoted extracellular matrix remodeling in breast cancer: LOXL2 upregulation and subsequent ECM alignment

Simion C. Dinca et al.

Summary: High co-expression of LOXL2 and OSM in IDC patients is associated with worse metastasis-free survival rates, while high levels of either individually are less impactful; LOXL2 expression is positively correlated with OSM/OSM receptor (OSMR) expression in IDC patients. Additionally, OSM-induced LOXL2 promotes an increase in ECM collagen I fiber crosslinking, leading to significant fiber alignment between cells and increased IDC cell invasion.
