Multi-omics of human plasma reveals molecular features of dysregulated inflammation and accelerated aging in schizophrenia

Anaamika Campeau et al.

Summary: Schizophrenia is a devastating psychiatric illness that is associated with reduced longevity and rapid aging. This study uses proteomics and metabolomics analyses to uncover molecular changes associated with schizophrenia over the human lifetime, particularly highlighting dysregulation of inflammatory and metabolic pathways. The findings provide insights into the molecular etiology of schizophrenia and its associated comorbidities throughout the aging process.

Altered Metabolism in Alzheimer Disease Brain: Role of Oxidative Stress

Nicole G. Rummel et al.

Summary: Alzheimer's disease is common among the aging population, but aging does not necessarily lead to neurodegeneration and memory decline. Recent research focuses on metabolic changes in the AD brain related to oxidative stress, with potential intervention targets identified. The main issue in AD is an imbalance of oxidative stress, particularly within the central nervous system, leading to disruptions in metabolic processes and cell damage. Understanding the role of reactive oxygen species in these alterations can potentially lead to therapeutic options for slowing the progression of AD.

Systematic Review and Meta-Analysis of Mass Spectrometry Proteomics Applied to Human Peripheral Fluids to Assess Potential Biomarkers of Schizophrenia

Joao E. Rodrigues et al.

Summary: Mass spectrometry (MS)-based proteomics can effectively identify biomarkers and relevant biological pathways in schizophrenia (SCZ) patients, providing potential targets for prediction and diagnosis improvement. This study identified 217 differentially expressed proteins in SCZ patients through meta-analysis of 19 articles using MS proteomics approaches. Enriched biological pathways included lipid metabolism, complement and coagulation cascades, and immune response. The upregulation of FCN3 and downregulation of APO1, APOA2, APOC1, and APOC3 were suggested as potential biomarkers for SCZ. However, heterogeneity in the findings and confounding factors should be taken into account for future studies to enhance the understanding of differentially expressed proteins.

BET-ting on histone proteomics in schizophrenia

Marija Kundakovic

Summary: In a recent study, Farrelly, Zheng and colleagues discovered an increase in histone variant H2A.Z acetylation associated with schizophrenia. They identified the BET protein BRD4 as an H2A.Z acetylation 'reader' and found that a BRD4 inhibitor could potentially treat schizophrenia.

Lower levels of tubulin alpha 1b in the frontal pole in schizophrenia supports a role for changed cytoskeletal dynamics in the aetiology of the disorder

Megan Snelleksz et al.

Summary: Lower levels of TUBA1B protein in the BA 10 region of patients with schizophrenia indicate a potential breakdown in cytoskeletal homoeostasis, unrelated to drug treatment effects.

Identification of bioactive metabolites in human iPSC-derived dopaminergic neurons with PARK2 mutation: Altered mitochondrial and energy metabolism

Justyna Okarmus et al.

Summary: PARK2 knockout cells exhibit increased TCA cycle activity, disturbed mitochondrial ultrastructure, ATP depletion, and dysregulation of glycolysis and carnitine metabolism. These perturbations are associated with elevated oxidative stress and reduced anti-oxidative response. Key findings were validated using patient-specific iPSC-derived neurons.
