High-speed super-resolution imaging of rotationally symmetric structures using SPEED microscopy and 2D-to-3D transformation

Summary: SPEED microscopy offers an effective approach to achieve 3D sub-diffraction-limit information in subcellular structures and organelles with rotational symmetry. It does not rely on complex optical components and can be implemented onto a standard inverted epifluorescence microscope, providing high spatiotemporal resolution for studying individual immobile or moving fluorescent molecules inside biological channels or cavities. The complete protocol takes around 9 days to complete, including cell culture and sample preparation, SPEED imaging, and data analysis and validation through simulation.
