Advances in Drugs Targeting Lymphangiogenesis for Preventing Tumor Progression and Metastasis

Chuqi Wang et al.

Summary: Metastasis of cancer cells through lymphatic vessels is a major cause of death in patients with malignancies. Recent studies have shown that tumor-induced lymphangiogenesis is closely related to lymphatic metastasis and poor clinical outcomes, providing new potential therapeutic targets for cancer treatment. This review provides an updated summary of the current advances in therapies preventing lymphatic metastasis and discusses the validity of different applications.

Engineering in vitro immune-competent tissue models for testing and evaluation of therapeutics

Jennifer H. Hammel et al.

Summary: Advancements in 3D cell culture, microscale fluidic control, and cellular analysis have allowed for the development of more physiologically-relevant engineered models of human organs. However, these models often overlook the important role of the immune system. This review discusses key components to consider in designing immune-competent models and reviews recent applications of immune models for cancer therapeutics screening, engineered T cell evaluation, autoimmune disease modeling, and vaccine efficacy screening.

Lymphangiogenesis in renal fibrosis arises from macrophages via VEGF-C/VEGFR3-dependent autophagy and polarization

Ying Zhang et al.

Summary: This study demonstrated a close relationship between lymphangiogenesis and renal fibrosis, with M1 macrophages predominately transdifferentiating into LECs. The VEGF-C/VEGFR3 signaling pathway was found to downregulate macrophage autophagy, promoting M1 macrophage polarization. These results suggest that M1 macrophages play a role in promoting lymphangiogenesis in renal fibrosis microenvironment, with VEGF-C/VEGFR3 pathway activating macrophage M1 polarization by suppressing macrophage autophagy.

Golnaz Anvari et al.

Summary: The study revealed that in obese adipose tissue, hypoxia leads to adipocyte hypertrophic expansion without new blood vessel formation, resulting in fibrosis. Using an in vitro model, it was found that hypoxia induces a diseased phenotype in adipocytes by regulating actin stress fiber formation, providing new potential therapeutic targets for obesity.
