Silk fibroin production in Escherichia coli is limited by a positive feedback loop between metabolic burden and toxicity stress

Novel insights into construct toxicity, strain optimization, and primary sequence design for producing recombinant silk fibroin and elastin-like peptide in E. coli

Summary: Spider silk proteins can be produced through recombinant production, but current difficulties include low yield, plasmid instability, and low translational efficiency. This study identified basal expression restriction and stress response-related genetic mutations as primary factors for increasing recombinant silk protein yield in E. coli. Additionally, a longer culturing time and attenuated induction in the bioprocess design, as well as targeted alteration in the primary sequence, were found to raise recombinant silk protein yield and mitigate toxicity. These findings provide multiple points of focus for further optimization of recombinant spider silk production.
