Zebrafish Tric-b is required for skeletal development and bone cells differentiation

Francesca Tonelli et al.

Summary: This study used zebrafish to investigate the role of TRIC-B in skeletal tissue. The researchers found that the tmem38b gene is expressed at early developmental stages in zebrafish, while tmem38a gene is not. Mutations in tmem38b resulted in under-modified collagen type I and intracellular retention. These findings contribute to our understanding of the importance of TRIC-B in bone cell differentiation.

Improvement of the skeletal phenotype in a mouse model of diastrophic dysplasia after postnatal treatment with N-acetylcysteine

Chiara Paganini et al.

Summary: Treatment with N-acetylcysteine (NAC) can improve cartilage proteoglycan sulfation and skeletal phenotype in a mouse model of diastrophic dysplasia (DTD). Prolonged treatment shows significant increase in sulfation and improvement in bone quality. The findings suggest that NAC may be an effective source of intracellular sulfate for DTD mice, potentially leading to a pharmacological treatment for DTD patients.

Knocking out TMEM38B in human foetal osteoblasts hFOB 1.19 by CRISPR/Cas9: A model for recessive OI type XIV

Laura Leoni et al.

Summary: Osteogenesis imperfecta (OI) type XIV is a rare bone disorder caused by mutations in the TMEM38B gene, leading to changes in the potassium channel TRIC-B in the endoplasmic reticulum membrane. Using CRISPR/Cas9 to knock out TMEM38B can provide a model for OI type XIV, showing altered cellular functions relevant to the disease. This technology is cost-effective and widely applicable for studying heritable disorders.
