Rapid Insights into the Stabilization of Interferon Alpha by Two Surfactants Revealed by STD-NMR Spectroscopy

Hristo L. Svilenov et al.

Summary: This study demonstrates the application of saturation transfer difference (STD) nuclear magnetic resonance (NMR) spectroscopy in detecting protein-surfactant interactions. It was found that polysorbate 20 binds with interferon alpha, with stronger binding at pH closer to the protein's iso-electric point. In contrast, no interactions were detected between poloxamer 407 and interferon alpha. This research highlights the potential of STD-NMR for quickly assessing surfactant-protein interactions and aiding in surfactant selection for protein formulation.

Summary: Solid-state storage is preferred for protein therapeutics to improve stability, with lyophilization as an effective drying method. However, the lyophilization process can lead to protein instability, which may be addressed by adding stabilizers or modifying the process for enhanced stability.

Toward Biotherapeutics Formulation Composition Engineering using Site-Identification by Ligand Competitive Saturation (SILCS)

Sandeep Somani et al.

Summary: The study introduces a novel in silico approach, SILCS-Biologics, for structure-based modeling of protein formulations, predicting potential hotspot regions on the protein surface for interactions among different components of a formulation. Experiments on a Fab domain of a monoclonal antibody showed that arginine increased viscosity, lysine reduced viscosity, and proline had no impact on viscosity. Efforts are now focused on further validating this computational framework and expanding its application to model full mAb and other protein therapeutics.

Summary: This study utilized SILCS methodology to predict protein-ligand binding affinities on eight target proteins, optimizing atomic free energy contributions using a Bayesian Markov chain Monte Carlo approach to improve prediction accuracy. The results validate the accuracy and computational efficiency of SILCS methodology for lead optimization and drug discovery.
