Reversibility and irreversibility in the temperature denaturation of monoclonal antibodies

Summary: This study found that the temperature denaturation of the CH2 domain of monoclonal antibodies is reversible, only becoming irreversible after denaturation of the Fab domain. A mixed denaturation model was developed to explain the experimental data and account for the pH dependence of the heat capacity function of 17b and other monoclonal antibodies. This more detailed analysis of monoclonal antibody stability is expected to contribute to the development of better approaches for understanding and optimizing the structural viability of therapeutic macromolecules.
