Cyclophilin D Regulates Oxidative Stress and Apoptosis via Mitochondrial Permeability Transition Pore in Acute Acalculous Cholecystitis

Kunqiao Hong et al.

Summary: Acute acalculous cholecystitis (AAC) is a disease with acute onset, rapid progression, high mortality, and various complications. Cyclophilin D (CypD) plays a proinflammatory role by regulating the opening of the mitochondrial permeability transition pore. Inhibiting the activity of CypD can reduce oxidative stress and apoptosis, thereby alleviating the symptoms of the disease.

Cellular and Molecular Signatures of Oxidative Stress in Bronchial Epithelial Cell Models Injured by Cigarette Smoke Extract

Chiara Cipollina et al.

Summary: Exposure of airway epithelium to environmental insults, such as cigarette smoke, leads to increased oxidative stress, which promotes the development of chronic lung diseases. Oxidative stress disrupts antioxidant defenses, affects cellular autophagy and survival regulatory mechanisms, leading to cell senescence. These events are exacerbated by inflammation driven by oxidative stress. Various models of bronchial epithelial cells are used to study the mechanisms and functions altered by cigarette smoke exposure and to test the efficacy of antioxidants. This review provides a comprehensive synthesis of in-vitro and ex-vivo studies from 2011 to 2021, investigating the molecular and cellular mechanisms triggered by cigarette smoke exposure in bronchial epithelial cells, commonly used experimental models, and the mechanisms of action of cellular antioxidant systems, as well as natural and synthetic antioxidant compounds.

Mitochondria as a Cellular Hub in Infection and Inflammation

Pauline Andrieux et al.

Summary: Mitochondria are the energy powerhouse of the cell, playing essential roles in inflammation, pathogen response, and oxidative stress. Pathogens can exploit mitochondria to disrupt cellular functions by affecting oxidative phosphorylation mechanisms. Targeting mitochondria could be a promising therapeutic strategy for infectious and inflammatory diseases.

The mitochondrial permeability transition pore: an evolving concept critical for cell life and death

Giampaolo Morciano et al.

Summary: This review summarizes the intriguing phenomenon of the mitochondrial permeability transition pore (mPTP) in cell biology. Despite almost 50 years of research, the mechanisms of mPTP are still not definitively understood. From initially being considered an in vitro artifact to now being recognized for its physiological and pathological implications, the journey of mPTP research has been long and complex.

MAPK15-ULK1 signaling regulates mitophagy of airway epithelial cell in chronic obstructive pulmonary disease

Ming Zhang et al.

Summary: In COPD patients, airway epithelial cells show increased phosphorylation of MAPK15 and ULK1, as well as enhanced expression of Beclin1 and the ratio of LC3II to LC3I. Cigarette smoke extract activates the MAPK15-ULK1 signaling pathway, leading to mitophagy and mitochondrial oxidative stress in airway epithelial cells. Targeting this pathway may be a potential therapeutic strategy for COPD.
