Global burden of heart failure: a comprehensive and updated review of epidemiology

Gianluigi Savarese et al.

Summary: Heart Failure (HF) is a multi-faceted and life-threatening syndrome with significant morbidity and mortality worldwide, affecting over 64 million people. While the incidence of HF has stabilized in industrialized countries, the prevalence is increasing due to population ageing and improved treatment. Developing countries lack substantial data on HF epidemiology, which shows different features compared to the Western world. This review provides updated estimates on global burden of HF including prevalence, incidence, outcomes, and costs.

Myocardin-related transcription factor A regulates integrin beta 2 transcription to promote macrophage infiltration and cardiac hypertrophy in mice

Li Liu et al.

Summary: Macrophage-specific deletion of MRTF-A can alleviate cardiac hypertrophy, reduce heart function loss, and decrease cardiac fibrosis. The study suggests that MRTF-A may regulate macrophage activity and contribute to the pathogenesis of cardiac hypertrophy by activating ITGB2 transcription.

Summary: Heart failure is a common complication of diabetes, even in the absence of other cardiovascular diseases. With the increasing prevalence of diabetes globally, particularly type 2 diabetes, the burden of heart failure on the healthcare system is expected to rise. This consensus report aims to provide clear guidance to practitioners on screening and diagnosing heart failure in individuals with diabetes.

Trans-activation of eotaxin-1 by Brg1 contributes to liver regeneration

Zhiwen Fan et al.

Summary: Infiltration of eosinophils is associated with and contributes to liver regeneration. Eotaxin-1 expression is elevated in proliferating murine livers and can be stimulated by hepatocyte growth factor (HGF). Brg1 is a chromatin remodeling protein that directly binds to the eotaxin promoter to activate its transcription, and interacts with NF-kappa B/RelA to activate eotaxin transcription. Depletion of eotaxin or Brg1 deficiency delays liver regeneration.

Mitochondrial damage and activation of the cytosolic DNA sensor cGAS-STING pathway lead to cardiac pyroptosis and hypertrophy in diabetic cardiomyopathy mice

Meiling Yan et al.

Summary: This study found that the cGAS-STING signaling pathway is activated in diabetic hearts, leading to the activation of the NLRP3 inflammasome and release of proinflammatory cytokines. Knockdown of STING can alleviate cardiac pyroptosis and inflammation, and prevent diabetes-induced cardiac hypertrophy.

Exercise training after myocardial infarction increases survival but does not prevent adverse left ventricle remodeling and dysfunction in high-fat diet fed mice

Carmem Peres Valgas Da Silva et al.

Summary: This study demonstrates that aerobic exercise can significantly improve survival after myocardial infarction (MI), independent of its effects on cardiac function. However, exercise does not have an impact on obesity and glucose intolerance induced by a high-fat diet (HFD). This highlights the importance of considering environmental interventions, including diet, in the cardiac rehabilitation of obese patients.

SIRT6 Mitigates Heart Failure With Preserved Ejection Fraction in Diabetes

Summary: The study reveals that reduced endothelial SIRT6 expression is associated with the development of diabetes-related HFpEF, and improving cardiac function by inhibiting endothelial fatty acid uptake. Activation of SIRT6 with medication reduces cardiac lipid accumulation and improves diastolic function.

The Chromatin Remodeling Protein BRG1 Regulates SREBP Maturation by Activating SCAP Transcription in Hepatocytes

Ming Kong et al.

Summary: BRG1 deficiency in mice results in reduced processing and nuclear accumulation of SREBP in the liver, affecting lipid metabolism. BRG1 plays a crucial role in regulating SREBP by activating SCAP transcription, ultimately influencing SREBP target gene expression.

Redox-sensitive activation of CCL7 by BRG1 in hepatocytes during liver injury

Ming Kong et al.

Summary: Liver injuries induced by various stimuli lead to an upregulation of CCL7 expression, which plays an important role in macrophage migration during liver injury. This process is regulated by BRG1, which interacts with AP-1 and is phosphorylated by CK2, and this pathway is sensitive to redox changes. This study provides new insights into the mechanisms of liver inflammation and repair.

Cardiac fibrosis

Nikolaos G. Frangogiannis

Summary: Myocardial fibrosis, characterized by the expansion of cardiac interstitium through deposition of extracellular matrix proteins, is a common pathophysiologic feature in various myocardial conditions. Activated fibroblasts and myofibroblasts play a central role in cardiac fibrosis, producing matrix proteins and triggering fibrogenic signalling cascades in response to stress. Immune cells, vascular cells, and cardiomyocytes can also contribute to fibrosis, while fibrotic changes may disrupt cardiac function and play a role in arrhythmogenesis.
