The Role of Orexin-1 Receptors Within the Hippocampal CA1 Area in the Extinction and Reinstatement of Methamphetamine-Seeking Behaviors

Ali Veisi et al.

Summary: This study investigated the role of orexin-1 receptors (OX1R) in the hippocampal CA1 subregion in the extinction and reinstatement of methamphetamine-induced conditioned place preference. The findings showed that blocking OX1R in the CA1 area during the extinction period reduced the extinction latency and maintenance of rewarding aspects of methamphetamine. Additionally, blocking OX1R on the reinstatement phase prevented methamphetamine-induced reinstatement of drug-seeking behaviors. These results provide further evidence for the involvement of hippocampal OX1R in the maintenance and relapse of methamphetamine addiction.

A wide range of Deep Brain Stimulation of the nucleus accumbens shell time independently reduces the extinction period and prevents the reinstatement of methamphetamine-seeking behavior in rats

Kiarash Eskandari et al.

Summary: Methamphetamine (METH) addiction is a major public health issue that is difficult to cure with standard therapies. Deep Brain Stimulation (DBS) has shown promise in treating addiction by modulating neural activity. This study investigated the therapeutic effects of high- or low-frequency stimulation of the nucleus accumbens shell (NAcSh) on the extinction and reinstatement of METH-conditioned place preference (CPP). The results demonstrated that both high- and low-frequency stimulation reduced the extinction period of METH-induced CPP and prevented reinstatement of drug-seeking behavior.

Summary: Deep brain stimulation (DBS) is a neurosurgical procedure that allows targeted circuit-based neuromodulation and is now commonly used for the treatment of movement disorders such as Parkinson's disease, tremor, and dystonia. Advances in engineering and imaging technologies are expected to enhance the efficacy and tolerability of DBS procedures in the future.

Summary: Deep brain stimulation (DBS) has shown promise as a therapeutic modality for intractable addiction, particularly when targeting structures within the reward circuit like the nucleus accumbens. Current clinical data suggests that DBS is safe in patients with addiction and that targeting the nucleus accumbens may be the most effective approach in treating addiction.

Subthalamic low-frequency oscillations predict vulnerability to cocaine addiction

Mickael Degoulet et al.

Summary: Identifying vulnerable individuals before they transition to compulsive drug seeking is a challenge in addiction prevention. Abnormal oscillatory activity in the STN predicts compulsive-like behavior, and STN DBS at 30 Hz reduces pathological cocaine seeking in compulsive individuals. Early electrical signatures of future compulsive behaviors can inform addiction treatment strategies.
