EVI1 drives leukemogenesis through aberrant ERG activation

Johannes Schmoellerl et al.

Summary: Chromosomal rearrangements involving the MDS1 and EVI1 complex locus (MECOM) on chromosome 3q26 define an aggressive subtype of acute myeloid leukemia (AML) that is associated with chemotherapy resistance and dismal prognosis. EVI1 controls the expression of ERG and occupies a conserved intragenic enhancer region in AML, and interfering with this regulatory axis may provide entry points for the development of rational targeted therapies.

Srebflc preserves hematopoietic stem cell function and survival as a switch of mitochondrial metabolism

Yukai Lu et al.

Summary: Srebf1c gene plays a crucial role in regulating the fate of hematopoietic stem cells by modulating the activity of the TSC1-mTOR-mitochondria axis to restrain excessive activation of mitochondrial metabolism.

The STAT3-MYC axis promotes survival of leukemia stem cells by regulating SLC1A5 and oxidative phosphorylation

Maria L. Amaya et al.

Summary: This study reveals that acute myeloid leukemia (AML) stem cells rely on oxidative phosphorylation (OXPHOS) for energy metabolism and survival, which is mediated by signal transducer and activator of transcription 3 (STAT3). STAT3 regulates AML-specific expression of MYC and SLC1A5, and inhibition of SLC1A5 acts similarly to STAT3 inhibition by impairing OXPHOS. Furthermore, a novel small molecule STAT3 inhibitor selectively induces cell death in AML stem cells while sparing normal hematopoietic cells.

CD63 acts as a functional marker in maintaining hematopoietic stem cell quiescence through supporting TGFβ signaling in mice

Mengjia Hu et al.

Summary: The study identifies CD63 as a novel functional marker of HSCs in mice, showing that high CD63 expression is associated with more quiescent HSCs with robust self-renewal and myeloid differentiation abilities. CD63 deficiency leads to reduced HSC numbers, impaired quiescence, and increased sensitivity to irradiation and 5-fluorouracil treatment. Further investigations reveal that CD63 interacts with TGF beta receptors to sustain TGF beta signaling activity and regulate HSC quiescence.

Role of c-Myc haploinsufficiency in the maintenance of HSCs in mice

Yue Sheng et al.

Summary: This study investigated the dosage effect of c-Myc on hematopoiesis and its role in the Wnt/beta-catenin pathway in HSC and bone marrow niche cells. The results demonstrated the critical role of Nr4a1, Nr4a2, and Jmjd3 as downstream targets of c-Myc in HSCs. Additionally, the study provided evidence of c-Myc-mediated Apc loss inducing IL6 secretion in endothelial cells and its impact on erythroid cell differentiation.

TWIST1 preserves hematopoietic stem cell function via the CACNA1B/Ca2+/mitochondria axis

Nan Wang et al.

Summary: The transcription factor TWIST1 is identified as a critical regulator of HSC maintenance by modulating mitochondrial function. Deletion of Twist1 leads to decreased HSC frequency, impaired self-renewal capacity, skewed myeloid differentiation, and compromised stress tolerance. The mechanism involves activation of voltage-gated calcium channel Cacna1b, resulting in increased mitochondrial calcium levels, metabolic activity, and reactive oxygen species production, which can be rescued by calcium channel blocker under stress conditions.

Hematopoietic versus leukemic stem cell quiescence: Challenges and therapeutic opportunities

Eimear O'Reilly et al.

Summary: Hematopoietic stem cells (HSC) are responsible for producing mature blood cells, with most HSCs being in a state of quiescence and controlled by intrinsic and extrinsic mechanisms. In acute myeloid leukemia (AML), leukemic cells take over the hematopoietic bone marrow niche, acquiring a quiescent state and resistance to chemotherapy. Efforts to target driver mutations in AML are progressing, but addressing drug resistance in quiescent leukemic stem cells (LSCs) remains a challenge.

A STAT5B-CD9 axis determines self-renewal in and leukemic stem cells

Sebastian Kollmann et al.

Summary: STAT5B plays a unique role in driving self-renewal of hematopoietic and leukemic stem cells, specifically activating genes associated with quiescence and self-renewal, including CD9. CD9 levels serve as a prognostic marker for patients with STAT5-driven leukemia, and anti-CD9 antibodies may be useful in targeting and eliminating leukemic stem cells. Consideration of STAT5A and STAT5B as distinct entities is crucial in normal and malignant hematopoiesis.
