A metabolomic signature of the APOE2 allele

Summary: By conducting a meta-analysis of five studies, this research identified a signature of 19 metabolites significantly associated with the e2 allele of the APOE gene. These metabolites were primarily glycerolipids and glycerophospholipids, which were higher in carriers of the e2 allele. Additionally, one organic acid metabolite was also found to be associated with the e2 allele. However, no significant associations were found with metabolites in the e4 allele group.

Physical resilience as a moderator of the relationship between frailty and disability in older adults with osteoarthritis

Summary: Frailty is a leading cause of deteriorating physical function in older adults with osteoarthritis. This study found that frailty mediates the relationship between osteoarthritic symptoms and disability, and physical resilience moderates both the direct and indirect pathways of this mediating relationship. These findings emphasize the detrimental effects of osteoarthritic symptoms on disability through frailty, and suggest that these effects are conditional on an individual's physical resilience.

The majority of the included articles involved older adults with fractures, cardiac conditions, and cancer. Measurement instruments mainly focused on body function or structure, activity and participation, but lacked instruments capturing the societal aspects of physical resilience.

Summary: Frailty is a common geriatric syndrome strongly associated with disability, mortality, and hospitalization, with a genetic basis suggested to have heritability estimates between 30 and 45%. This study identified 14 loci associated with the frailty index, with an estimated single nucleotide polymorphism (SNP) heritability of 11%, and a higher educational attainment genetic risk score was linked to lower levels of frailty.

Trajectories of frailty in aging: Prospective cohort study

Summary: The study identified four distinct frailty trajectories in aging, including stable, mild, moderate, and severely frail trajectories. The worst trajectories were associated with modifiable risk factors such as low education, living alone, obesity, and physical inactivity. Proteomic analysis revealed proteins in lipid metabolism and growth factor pathways associated with frailty trajectories. High mortality risks were observed in the mild, moderate, and severely frail trajectories compared to the stable group.
