Vector Affinity and Receptor Distribution Define Tissue-Specific Targeting in an Engineered AAV Capsid

An evolved AAV variant enables efficient genetic engineering of murine T cells

Summary: The precise targeting of large transgenes to T cells using homology-directed repair has been significant for adoptive cell therapies and T cell biology. The use of adeno-associated virus (AAV) for delivery of DNA templates has improved knockin efficiencies, but current AAV serotypes have limited tropism in human T cells used in immunodeficient mouse models. To overcome this, Ark313, a synthetic AAV that exhibits high transduction efficiency in murine T cells, was developed. Ark313 enables targeted knockins in murine T cells, allowing for preclinical modeling of Trac-targeted CAR-T and transgenic TCR-T cells in immunocompetent models. Efficient gene targeting in murine T cells holds great potential for improved cell therapies and advances in experimental T cell immunology.

Genetic Modification of the AAV5 Capsid with Lysine Residues Results in a Lung-Tropic Liver-Detargeted Gene Transfer Vector

Summary: The intravenous administration of adeno-associated virus (AAV) vectors can lead to gene expression in liver hepatocytes, but AAV capsids targeting the lung may be more effective for non-liver-based disease therapy. In this study, the researchers inserted positively charged lysine residues into the AAV5 capsid to enhance its distribution to the lung after intravenous administration. The modified AAV5 capsid, called AAV5-PK2, showed the highest biodistribution in the lung and significantly decreased distribution to the liver compared to AAV5. This lung-tropic AAV vector has the potential to be a valuable tool for lung-directed gene therapies.
