Profiling of syngeneic mouse HCC tumor models as a framework to understand anti-PD-1 sensitive tumor microenvironments

Daniel J. Zabransky et al.

Summary: The response to immune checkpoint inhibitors in hepatocellular carcinoma (HCC) varies among patients. To better understand the mechanisms of response and resistance, syngeneic mouse models are used to study the complex interactions between cancer cells and nonmalignant cells in the tumor microenvironment. These models reveal the heterogeneity of immune tumor microenvironments and the existence of immunosuppressive pathways. Different models and human HCCs have diverse immune tumor microenvironments, highlighting the importance of using multiple syngeneic HCC models for immune modulation research.

Summary: This experiment evaluates the role of tumor-infiltrating lymphocytes (TILs) in distant tumors after cryoablation of breast cancer, and finds that cryoablation can induce a tumor-specific TIL response, preventing cancer recurrence and metastasis.

Cowpea Mosaic Virus and Natural Killer Cell Agonism for In Situ Cancer Vaccination

Edward C. Koellhoffer et al.

Summary: The plant virus CPMV has been shown to be an effective in situ cancer vaccine and recruit NK cells within the tumor microenvironment. In this study, a combination of CPMV and anti-41BB monoclonal antibody agonist was found to improve NK cell function and enhance the efficacy of in situ cancer vaccination. This dual therapy approach provided a robust antitumor response and improved survival in murine models of colon carcinomatosis and melanoma.
