Modeling of Fabry disease nephropathy using patient derived human induced pluripotent stem cells and kidney organoid system

Reduced α-galactosidase A activity in zebrafish (Danio rerio) mirrors distinct features of Fabry nephropathy phenotype

Summary: Fabry disease (FD) is a rare genetic lysosomal storage disorder caused by deficiency of alpha-galactosidase A (alpha-GAL) enzyme, leading to accumulation of glycosphingolipids in tissues. A zebrafish model with decreased expression of GLA gene was generated using CRISPR/Cas9 technology, showing similar symptoms to human FD such as increased plasma creatinine levels and proteinuria. This zebrafish model could be useful for studying novel biomarkers and innovative therapeutic approaches for FD.

Determining epigenetic memory in kidney proximal tubule cell derived induced pluripotent stem cells using a quadruple transgenic reprogrammable mouse

Summary: In this study, researchers reprogrammed PTC and TTF cells from a reprogrammable mouse to iPSCs and differentiated them to renal progenitors to investigate the role of epigenetic memory in renal differentiation. The results showed that there were no significant differences in gene expression and global epigenetic marks between PTC iPSCs and TTF iPSCs. Although PTC iPSC-derived renal cells expressed higher levels of three renal progenitor genes, there was no evidence of an epigenetic memory basis for this.

Human kidney organoids reveal the role of glutathione in Fabry disease

Summary: Fabry disease is a rare inherited disorder caused by a mutation in the GLA gene, leading to the accumulation of fat in cells and causing severe complications. By using kidney organoids with the GLA mutation, researchers found high oxidative stress levels and decreased metabolism of the antioxidant glutathione. Boosting glutathione improved cellular structures and reduced oxidative stress in the organoids.

Summary: Novel patient-specific human-induced pluripotent stem cell lines were generated from a female and a male with Fabry disease, both containing the same frameshift deletion mutation in the alpha-galactosidase A gene. These hiPSCs exhibited typical human embryonic stem cell-like morphology, expressed pluripotency-associated markers, and could differentiate into all three germ-layers. Karyotyping confirmed normal chromosome numbers for both patients. This study may offer additional insights into the pathophysiology of Fabry disease.

Modeling of endothelial cell dysfunction using human induced pluripotent stem cells derived from patients with end-stage renal disease

Fabry disease exacerbates renal interstitial fibrosis after unilateral ureteral obstruction via impaired autophagy and enhanced apoptosis

Summary: The study investigated the pathogenic mechanism leading to fibrosis in kidneys of Fabry disease mice with unilateral ureteral obstruction (UUO). The results showed an increased presence of lamellar inclusion bodies and fibrosis in the kidneys of diseased mice. Impaired autophagy and enhanced apoptosis were identified as probable mechanisms involved in the enhanced renal fibrosis observed under the stimulus of UUO in Fabry disease.
