Green synthesis of lignin-based nanoparticles as a bio-carrier for targeted delivery in cancer therapy

Khushboo Pathania et al.

Summary: Polymeric magnetic nanoparticles based on lignin have been developed for targeted delivery of methotrexate, showing higher efficacy in cancer diagnosis and treatment. The synthesized nanoparticles exhibit magnetic and smooth polyhedral traits with a dimension of 110-130 nm. The drug-loaded nanoparticles have high cytotoxicity in breast cancer and macrophage cell lines and exhibit sustained release of methotrexate.

Lignin-chitosan biocomposite film for antimicrobial activity: Fabrication, characterization and in-vitro evaluation

Pritiman Pothal et al.

Summary: In this study, lignin-chitosan polymeric films were prepared and characterized. The films exhibited smooth and uniform surface with fibrous structure, and showed no hemolytic activity and non-toxicity towards fibroblast cells. Furthermore, the drug-loaded film showed significant antimicrobial activity against both Gram-negative and Gram-positive bacteria.

Summary: This study delineates the formulation of poly(lactide-co-glycolic acid) nanoparticles (PLGA NPs) for drug encapsulation using nanoprecipitation method. Specifically, the encapsulation of immunomodulating agents BBIQ and Try in the PLGA NPs was performed to overcome their shortcomings. Various characterization techniques were employed to confirm the successful loading of the drugs and study their release behavior from the NPs.

Generation of potent cellular and humoral immunity against SARS-CoV-2 antigens via conjugation to a polymeric glyco-adjuvant

Laura T. Gray et al.

Summary: Spike-p(Man-TLR7) vaccination induces robust antigen-specific cellular and humoral responses in mice, generating high and long-lasting titers of antibodies with neutralizing abilities surpassing convalescent human serum. The vaccine also enhances germinal center B cells, antigen-specific antibody secreting cells, activated T follicular helper cells, and polyfunctional Th1-cytokine producing CD4(+) and CD8(+) T cells.

Prolonged Codelivery of Hemagglutinin and a TLR7/8 Agonist in a Supramolecular Polymer-Nanoparticle Hydrogel Enhances Potency and Breadth of Influenza Vaccination

Gillie A. Roth et al.

Summary: The sustained release of vaccine cargo has shown to improve humoral immune responses to challenging pathogens like influenza. This study introduces an injectable polymer-nanoparticle hydrogel system that prolongs the co-delivery of antigen and adjuvant, resulting in increased potency and durability of influenza subunit vaccines.

Evolution of Toll-like receptor 7/8 agonist therapeutics and their delivery approaches: From antiviral formulations to vaccine adjuvants

Sachin Bhagchandani et al.

Summary: Imidazoquinoline derivatives function as synthetic agonists of Toll-like receptors 7 and 8, with potential therapeutic utility in antitumor immunomodulation and vaccine adjuvants. However, challenges such as broad expression profiles, poor pharmacokinetic properties, and associated toxicities hinder their successful clinical translation. Strategies to enhance safety and efficacy through bioconjugates and nanoparticle formulations are being explored to address these barriers.
