In-Depth Characterization of Acidic Variants Induced by Metal- Catalyzed Oxidation in a Recombinant Monoclonal Antibody

Summary: Characterization of antibody charge heterogeneity is crucial in antibody drug development. This study presents a new workflow combining untargeted and targeted analyses to identify and characterize the induced acidic variants in a highly oxidized IgG1 antibody. The study identified 28 site-specific oxidation products as the sources of the induced charge heterogeneity and provides new insights into understanding the acidic charge heterogeneity of antibody drugs. The characterization workflow presented in this study can be widely applied as a platform approach in the biotechnology industry for in-depth characterization of antibody charge variants.

Direct coupling of size exclusion chromatography and mass spectrometry for the characterization of complex monoclonal antibody products
