Expert Algorithm for Substance Identification Using Mass Spectrometry: Statistical Foundations in Unimolecular Reaction Rate Theory

Glen P. P. Jackson et al.

Summary: This study aims to accurately identify an organic substance from its mass spectrum without analyzing a contemporaneous spectrum of the suspected substance. The first part of the report describes how a theoretical model predicts linear correlations between branching ratios in mass spectra under changing analysis conditions. The model was tested using a dataset of cocaine spectra, achieving accurate predictions even with significant variations in measured abundances. The second part of the report demonstrates how the model can be applied for reliable identification of cocaine and its diastereomers from other known negative spectra.

Incorporating measurement variability when comparing sets of high-resolution mass spectra

Matthew J. Roberts et al.

Summary: Mass spectra are important for identifying compounds. A mathematical approach incorporating measurement variability in comparing high-resolution mass spectra is presented. Consensus mass spectra representing each analyte in the comparison are constructed using replicate mass spectra, and the similarity between these data structures is computed. The approach is discussed in the context of discriminating methamphetamine and phentermine using in-source collision induced dissociation mass spectra collected with direct analysis in real time mass spectrometry.

Comparison of Cosine, Modified Cosine, and Neutral Loss Based Spectrum Alignment For Discovery of Structurally Related Molecules

Wout Bittremieux et al.

Summary: A comparative analysis was performed to evaluate the scoring properties of neutral loss matching, cosine similarity, and modified cosine similarity in MS/MS data. The results showed that the modified cosine similarity outperformed the other methods in all cases, and the performance of MS/MS spectrum alignment depended on the location, type, and chemical compound class of modifications.

Assessment of the similarity between in-source collision-induced dissociation (IS-CID) fragment ion spectra and tandem mass spectrometry (MS/MS) product ion spectra for seized drug identifications

Jared Sharp et al.

Summary: This study assesses the similarity between IS-CID fragment ion spectra and MS/MS product ion spectra through visual and statistical methods, as well as the repeatability of IS-CID fragment ion spectra. The results demonstrate the comparability of IS-CID fragment ion spectra and MS/MS product ion spectra for representative NPS, and the high similarity within a specific range of comparison.

Development of a Data Analysis Tool to Determine the Measurement Variability of Consensus Mass Spectra

Benjamin J. Place

Summary: The study presents a novel tool for calculating the variability of measured mass spectra, focusing on PFOA in different sample types. It found that the variability of measured ddMS2 for PFOA is similar in solution and house dust extract, with selected parameters varying between the sample types. The variability of ddMS2 spectra for PFOA in both samples was then used to calculate a more robust similarity factor for identifying unknown compounds.

The differentiation of synthetic cathinone isomers using GC-EI-MS and multivariate analysis

Ruby E. Liliedahl et al.

Summary: This study developed a canonical discriminant analysis approach for differentiating synthetic cathinone isomers, reducing the required number of sample injections. The research found that using multiple scans and reducing the number of ions can effectively distinguish different cathinone isomers, improving the classification rates.

A framework for the development of targeted gas chromatography mass spectrometry (GC-MS) methods: Synthetic cannabinoids

Edward Sisco et al.

Summary: This paper presents a six-step data-driven framework for developing and evaluating gas chromatography mass spectrometry (GC-MS) methods for targeted classes of drugs, focusing on maximizing retention time differences and understanding the trade-offs between sensitivity and reproducibility. By expanding the panel of compounds analyzed, limitations in compound discrimination were identified, and a targeted method for synthetic cannabinoids was successfully developed. Compared to a general method, the new targeted method showed significantly increased sensitivity, shorter analysis time, and major improvements in retention time differences.
