Enantioenriched BCP Benzylamine Synthesis via Metal Hydride Hydrogen Atom Transfer/Sulfinimine Addition to [1.1.1]Propellane

Russell Shelp et al.

Summary: This study reports a stereoselective synthesis method for BCP benzylamine derivatives using metal hydride hydrogen atom transfer, with high diastereoselectivity. The streamlined synthesis of a key drug intermediate BCP analogue demonstrates the strategic impact of the method. The mechanistic evidence for a competitive H-2 evolution pathway and the importance of controlled silane addition during reaction initiation are disclosed.

Dicarbofunctionalization of [1.1.1]Propellane Enabled by Nickel/Photoredox Dual Catalysis: One-Step Multicomponent Strategy for the Synthesis of BCP-Aryl Derivatives

Weichen Huang et al.

Summary: Bicyclo[1.1.1]pentane (BCP) motifs have become important in pharmaceutical, agrochemical, and materials chemistry. Most of these structures are obtained through the cleavage of the internal C-C bond in [1.1.1]propellane, using either radicals or metal-based nucleophiles. A multicomponent reaction that allows direct access to complex and diverse disubstituted BCP products is more attractive than current stepwise approaches.
