An NmrA-like enzyme-catalysed redox-mediated Diels-Alder cycloaddition with anti-selectivity

Summary: The Diels-Alder cycloaddition is a powerful method in organic synthesis, which is commonly used to synthesize important pharmaceuticals. However, controlling the stereoselectivity of these reactions is challenging, and new strategies are needed to construct complex molecules with desired chirality. In this study, a new bifunctional oxidoreductase/Diels-Alderase enzyme called CtdP was discovered and characterized. It was found to catalyze the inherently disfavoured cycloaddition with strict alpha-anti-selectivity, and the redox mechanism for this reaction was revealed.

YcaO-mediated ATP-dependent peptidase activity in ribosomal peptide biosynthesis

Summary: YcaO enzymes catalyze ATP-dependent post-translation modifications on peptides, including the installation of (ox/thi)azoline, thioamide and/or amidine moieties. In this study, we found that the YcaO enzyme MusD in the biosynthesis of muscoride A performs both ATP-dependent cyclodehydration and peptide bond cleavage, which is a novel mechanism. Through structural and biochemical analysis, we identified an acyl-phosphate species consistent with the proposed mechanism for backbone amide activation. Bioinformatics analysis also revealed other cyanobactin pathways that may use bifunctional YcaO enzymes. These studies expand the understanding of the modifications catalyzed by YcaO proteins and provide evidence for a unifying mechanism.

Engineering C-C Bond Cleavage Activity into a P450 Monooxygenase Enzyme

Summary: The study focused on the carbon-carbon cleavage reaction of alpha-hydroxy ketones catalyzed by cytochrome P450 enzymes. It was found that an engineered CYP199A4 enzyme can facilitate this reaction, showing an inverse solvent isotope effect similar to the lyase activity of CYP17A1. Structural analysis revealed that the substrate binds to the active site of the enzyme in a specific conformation, mimicking the lyase reaction in CYP17A1. This study opens up new possibilities for further characterization of CYP catalytic intermediates.

Set of Cytochrome P450s Cooperatively Catalyzes the Synthesis of a Highly Oxidized and Rearranged Diterpene-Class Sordarinane Architecture

Summary: Cytochrome P450s are versatile oxidases that catalyze important chemical transformations in the biosynthesis of natural products. We discovered a group of P450s that can reshape the cycloaraneosene framework to form a sordarinane architecture. One of these enzymes, SdnB, cleaves the C-C bond of the epoxy residue to yield formyl groups, while SdnF selectively oxidizes one of the formyl groups to a carboxyl group and accelerates the final cyclization reaction.

Classes of Amides that Undergo Selective N-C Amide Bond Activation: The Emergence of Ground-State Destabilization

Summary: Ground-state destabilization of the N-C(O) linkage is a powerful tool to functionalize traditionally inert amide bonds. This Perspective summarizes the contributions of our laboratory in the development of new ground-state-destabilized amide precursors and their synthetic utility in cross-coupling and acyl addition reactions. Ground-state-destabilized amides can be used as electrophiles to forge new C-C, C-N, C-O, C-S, C-P, and C-B bonds, enabling a plethora of previously unknown transformations of the amide bond.
