Coordination Assistance: A Powerful Strategy for Metal-Catalyzed Regio- and Enantioselective Hydroalkynylation of Internal Alkenes

Summary: Alkenes are versatile compounds that can be obtained from industry or synthesized organically. The development of catalytic asymmetric alkene hydrofunctionalizations is driven by the widespread occurrence of alkenes, allowing for the construction of complex chiral molecules. By using a coordinating group on the alkene substrate, reactivity can be enhanced and the control of regio-, diastereo-, and enantioselectivities can be facilitated. Through this strategy, several alkene hydrofunctionalization methods with excellent controls have been developed.

Summary: In this study, a new method for achieving enantioselective [4+3] cyclization through C(sp(2))-H activation is described. A cooperative catalytic system consisting of a Cp*Rh(III) complex and a chiral Lewis base is used to successfully generate a variety of benzolactam products. The results demonstrate the importance of chiral Lewis base catalysis in controlling the enantioselectivity of transition metal-catalyzed C-H functionalizations.

Mechanistic Investigations of the Asymmetric Hydrogenation of Enamides with Neutral Bis(phosphine) Cobalt Precatalysts

Summary: The mechanism of asymmetric hydrogenation of prochiral enamides by well-defined cobalt(0) and cobalt(II) precatalysts using a chiral bis(phosphine) ligand has been investigated. Characterization of the resulting complexes and monitoring of the reactions revealed the resting state of the catalyst, the rate law, and the irreversible H2 addition step. Computational and labeling studies provided evidence for a two-electron redox cycling during the catalytic cycle. The experiments support an unsaturated pathway for the hydrogenation of prochiral enamides.

Advancing the Logic of Chemical Synthesis: C-H Activation as Strategic and Tactical Disconnections for C-C Bond Construction

Summary: The design of synthetic routes by retrosynthetic logic is influenced by available transformations. Transition-metal-catalyzed C-H activation is a powerful strategy for C-C bond formation, but its uptake in total synthesis has been tepid due to synthetic intractability and a lack of comprehensive guidelines. This Review addresses these issues and offers a guide to identify retrosynthetic opportunities to generate C-C bonds by C-H activation processes. Comparing traditional approaches and recent C-H activation methods, this Review demonstrates more efficient retrosynthetic strategies enabled by C-H activation for C-C bond construction.

Cp*Co(III)-Catalyzed Enantioselective Hydroarylation of Unactivated Terminal Alkenes via C-H Activation

Summary: The study reports a Cp*Co(III)-catalyzed asymmetric hydroarylation of unactivated aliphatic terminal alkenes assisted by tailor-made amino acid ligands. A novel noncovalent interaction was found critical for the chiral induction, resulting in high yields and excellent enantioselectivities of C2-alkylated indoles. DFT calculations revealed the reaction mechanism and origins of chiral induction in the stereodetermining alkene insertion step.

Combined Theoretical and Experimental Studies Unravel Multiple Pathways to Convergent Asymmetric Hydrogenation of Enamides

Mechanistic studies revealed different pathways for enantioconvergence, including fast isomerization for aryl enamides and substrate chelation for alkyl enamides. DFT calculations were performed to predict the correct absolute configuration of the products and strengthen the proposed mechanism of the iridium-catalyzed isomerization pathway.
