Site- and enantioselective cross-coupling of saturated N-heterocycles with carboxylic acids by cooperative Ni/photoredox catalysis

Summary: Site- and enantioselective alpha-acylation of saturated N-heterocycles with in situ-activated carboxylic acids is achieved using a modular approach that combines hydrogen-atom-transfer reactivity of photocatalytically generated chlorine radicals with asymmetric nickel catalysis. This strategy selectively functionalizes cyclic alpha-amino C-H bonds without affecting benzylic, allylic, acyclic alpha-amino, and alpha-oxy methylene groups. The protocol displays excellent chemo-, site-, and enantioselectivity and can be used for late-stage diversification, including the synthesis of previously inaccessible Taxol derivatives.

Catalyst-Controlled Regiodivergent and Enantioselective Formal Hydroamination of N,N-Disubstituted Acrylamides to α-Tertiary-α-Aminolactam and β-Aminoamide Derivatives

Summary: We developed a catalyst-controlled regiodivergent and enantioselective formal hydroamination method for the synthesis of enantioenriched alpha-tertiary-alpha-aminolactam and alpha-chiral-beta-aminoamide derivatives. Sterically- and electronically-disfavored enantioselective hydroaminations of electron-deficient alkenes were successfully achieved by tuning different transition metals and chiral ligands. Furthermore, extremely hindered aliphatic alpha-tertiary-alpha-aminolactam derivatives were synthesized using a Cu-H catalyzed asymmetric C-N bond formation, while Ni-H catalyzed anti-Markovnikov-selective formal hydroaminations of alkenes provided access to enantioenriched alpha-chiral-beta-aminoamide derivatives. These reactions demonstrated good yields with high levels of enantioselectivity and tolerated a wide range of functional groups.
