Qualitative and quantitative evaluation of thylakoid complexes separated by Blue Native PAGE

Summary: In this study, the Blue/Clear-Native polyacrylamide gel electrophoresis (BN PAGE) method was improved by using a detergent mixture of 1% (w/V) n-dodecyl-beta-d-maltoside plus 1% (w/V) digitonin for solubilisation and 4.3-8% gel gradients for separation. Several large photosystem (PS) I containing bands were detected, and it was found that these bands were PSI-NADH dehydrogenase-like megacomplexes in maize bundle sheath thylakoids and PSI complexes with different light-harvesting complex (LHC) complements (PSI-LHCII, PSI-LHCII*) in mesophyll thylakoids of lincomycin treated maize. Proposed methods for quantitative determination and comparison of the complexes in different samples were also provided.

Long-term adaptation of Arabidopsis thaliana to far-red light

Summary: Vascular plants predominantly use carotenoids and chlorophylls a and b for solar energy absorption, with limited utilization of far-red (FR) light, unlike some cyanobacteria which have evolved to harness FR light through redesigning photosynthetic apparatus. Implementing similar strategies in plants to boost crop yield requires a better understanding of FR light-induced changes. Research on Arabidopsis thaliana exposed to FR light revealed adjustments in photosystem antenna size for energy input balance, leading to photoinhibition upon re-exposure to light. Notably, the alterations in photosynthetic membrane structure induced by FR light are not due to senescence, but rather a response to excitation imbalance between photosystems.

Comparative proteomics of thylakoids from Arabidopsis grown in laboratory and field conditions

Summary: In this study, quantitative label-free proteomics was used to show that field-grown Arabidopsis plants exhibit characteristics of both low and high light acclimation strategies observed in laboratory-grown plants. This indicates that plants in natural conditions undergo specific adaptations at the proteome level to optimize photosynthesis efficiency and cope with environmental stresses.
