Three-Fluorophore FRET Enables the Analysis of Ternary Protein Association in Living Plant Cells

Summary: Protein-protein interaction studies are crucial for understanding cellular signaling. This study used three-fluorophore FRET and FRET-fluorescence lifetime imaging microscopy techniques to demonstrate the formation of a ternary complex involving Receptor-Like Protein 44, Brassinosteroid Insensitive 1, and Brassinosteroid Insensitive 1 Associated Kinase 1 in living plant cells. The researchers also found that this complex is localized in a distinct plasma membrane nanodomain.

Summary: The Whispering gallery mode (WGM) microcavities are potential candidates in biosensing applications as their resonance wavelengths shift with changes in the refractive index. They offer high-quality resonance modes and accessible surface functionalities, making them promising for molecular assays, but their high sensitivity also makes them inherently unstable. The WGM resonances are also shown to strongly enhance fluorescence energy transfer between donors and acceptors, providing a potentially powerful class of biosensors combining sensitivity and simple detection schemes.

Summary: By interacting with the vacuum electromagnetic field in an optical microcavity, it is possible to control the tautomerization of single phthalocyanine molecules and alter their photoinduced tautomerization. Inside the microcavity, the average fluorescence lifetime of molecules decreases significantly, reducing the possibility of molecules entering the triplet state.
