Excluded Volume and Weak Interactions in Crowded Solutions Modulate Conformations and RNA Binding of an Intrinsically Disordered Tail

Resolving the enthalpy of protein stabilization by macromolecular crowding

Summary: Proteins in crowded cellular environments can affect their folded state stability, but most experiments are conducted in dilute solutions. Using F-19 nuclear magnetic resonance spectroscopy, we studied the unfolding thermodynamics of a protein domain in the presence of polyethylene glycols (PEGs) of different sizes and concentrations. Contrary to current theories, our experiments show that PEG stabilization is accompanied by heat release and entropy disfavors folding. We propose that different mechanisms drive stabilization by small and large PEGs.

The Analytical Flory Random Coil Is a Simple-to-Use Reference Model for Unfolded and Disordered Proteins

Summary: Unfolded proteins, including denatured, unfolded, and intrinsically disordered proteins, can be described using analytical polymer models. The analytical Flory random coil (AFRC) model, parameterized using all-atom simulations and polymer scaling theory, provides a simple-to-use reference state for comparing and normalizing experimental or computational results. The AFRC model allows identification of sequence-specific intramolecular interactions and contextualization of radii of gyration from small-angle X-ray scattering experiments of disordered proteins.

An in Vitro Cytomimetic of In-Cell RNA Folding

Summary: This study investigates the effects of cytoplasmic crowding and sticking on RNA stability using artificial crowding and sticking agents. The results show that the addition of these agents leads to stabilization and destabilization of RNA folding through excluded volume effect and surface interactions. By comparing folding stability inside living cells and in vitro, it is found that the cytoplasmic environment affecting RNA folding is similar to a combination of 20% v/v M-PERTM and 150 g/L PEG10k.

Macromolecular Crowding and Intrinsically Disordered Proteins: A Polymer Physics Perspective
