Role of Crystal Disorder and Mechanoactivation in Solid-State Stability of Pharmaceuticals

This review examines the existing literature on crystal disorder and its impact on solid-state stability of drugs, and discusses the generation, evolution, and measurement of crystal disorder. It also proposes a workflow to investigate the relevance of crystal disorder on the degradation of pharmaceuticals.

Oxidation of Drugs during Drug Product Development: Problems and Solutions

Summary: Oxidation is a common degradation pathway for pharmaceuticals, and it is more complex than hydrolysis, producing a wide range of degradation products. Accelerated studies of drug-excipient mixtures can provide realistic insights into degradation in the solid state, as excipients are the main sources of impurities that can initiate oxidation. Based on the results of these studies, appropriate measures can be taken to minimize the oxidation of solid drug products.

An empirical model for sorption by glassy polymers: An assessment of thermodynamic parameters

Through the analysis, the solvent-induced glass transition temperature variation can be estimated from the sorption isotherm.
