Antibodies to watch in 2023

Helene Kaplon et al.

Summary: In this article, the key events in the development of commercial monoclonal antibody therapeutics in 2022 are discussed, and events that may occur in 2023 are forecasted. By mid-November, 12 antibody therapeutics had received first approvals in the United States or European Union, and an additional 7 had been approved in China or Japan. Globally, at least 24 investigational antibody therapeutics are undergoing regulatory review. The data show that, excluding antibodies for COVID-19, the late-stage commercial clinical pipeline has grown by approximately 20% in the past year.

Modeling study of long-term stability of the monoclonal antibody infliximab and biosimilars using liquid-chromatography-tandem mass spectrometry and size-exclusion chromatography-multi-angle light scattering

Pauline Legrand et al.

Summary: This study implemented the accelerated stability assessment program (ASAP) to model the long-term stability of monoclonal antibodies (mAbs). Using LC-MS/MS and SEC-MALS-UV/RI analysis, post-translational modifications and degradation pathways of mAbs were characterized. Comparison of ASAP models and long-term stability data indicated the stability of mAbs. The study demonstrated the particularities of ASAP modeling for mAbs and its ability to accurately predict long-term stability.

Simultaneous quantification and structural characterization of monoclonal antibodies after administration using capillary zone electrophoresis-tandem mass spectrometry

Tessa Reinert et al.

Summary: Monoclonal antibodies have shown great success in therapeutic areas such as oncology and immune disorders. However, there is limited understanding of their structural evolution after administration, and significant inter-patient differences in clearance and clinical responses have been observed.
