A fully validated, selective, and sensitive bioanalytical method has been developed for the rapid quantification of these components. The oral pharmacokinetic profile of the four bioactive components in rats was evaluated using noncompartmental analysis and Phoenix WinNonlin software.

Preclinical pharmacokinetics of 4-hydroxy isoleucine using LC-MS/MS: a potential polycystic ovary syndrome phytopharmaceutical therapeutics

Summary: The aim of this study was to investigate the preclinical pharmacokinetics of 4-hydroxy isoleucine (4-HIL) targeted for polycystic ovary syndrome. LC-MS/MS was used for quantitative bioanalysis of 4-HIL in different biological matrices in female Sprage-Dawley rats. Results showed that at a dose of 50 mg/kg, 4-HIL had an absolute oral bioavailability of 56.8%. It was rapidly absorbed and distributed in various tissues, with the highest concentrations found in the small intestine, kidney, and ovary. Furthermore, 11.07% of 4-HIL was excreted in urine and feces within 72 hours. Based on tissue distribution results, 4-HIL levels in vital organs were deemed safe. Therefore, 4-HIL holds promise as a therapeutic option for managing polycystic ovary syndrome.
