High-fat diet-induced metabolic syndrome increases ligature-induced alveolar bone loss in mice

Zhongyang Lu et al.

Summary: This study found that high-fat diet-induced metabolic syndrome increases alveolar bone loss in mice with ligature-induced periodontitis, and palmitic acid and lipopolysaccharide cooperatively stimulate osteoclast formation and proinflammatory gene expression in macrophages.

Exercise as Treatment for Neuropathy in the Setting of Diabetes and Pre-diabetic Metabolic Syndrome: A Review of Animal Models and Human Trials

J. Robinson Singleton et al.

Summary: Exercise has shown promising potential as an alternative treatment for neuropathy in metabolic syndrome, with mechanisms including reducing inflammation and improving oxidative metabolism. Studies in animal models suggest beneficial effects of exercise on various aspects of neuropathy pathogenesis, while human studies indicate improvements in nerve regenerative capacity, pain, and functional performance measures. However, patient adherence remains a key limitation to realizing the full benefits of exercise therapy.

GPR40 Agonism Modulates Inflammatory Reactions in Vascular Endothelial Cells

Joo Won Kim et al.

Summary: Endothelial dysfunction is closely related to inflammatory responses, and G protein-coupled receptor 40 (GPR40) plays an important role in modulating inflammatory reactions in vascular endothelial cells. In this study, a specific agonist of GPR40, LY2922470, was found to inhibit the inflammatory responses induced by lipopolysaccharide (LPS), while another agonist, TAK875, had no effect. LY2922470 also reduced the expression of adhesion molecules and the attachment of cells. These findings suggest that GPR40 could be a potential therapeutic target for treating endothelial inflammation.

Loss of GPR40 in LDL receptor-deficient mice exacerbates high-fat diet-induced hyperlipidemia and nonalcoholic steatohepatitis

Zhongyang Lu et al.

Summary: This study demonstrated for the first time that loss of GPR40 in LDLR-deficient mice exacerbated HFD-induced hyperlipidemia, hepatic steatosis, inflammation, and fibrosis potentially through a CD36-dependent mechanism, suggesting that GPR40 may play a beneficial role in hyperlipidemia-associated NASH in LDLR-deficient mice.

Milagros Rojas et al.

Summary: Alzheimer's disease (AD) and type 2 diabetes mellitus (T2DM) are highly prevalent worldwide, especially among elderly individuals, and share various pathophysiological mechanisms, including alterations in insulin signaling and mitochondrial dysfunctions in the brain. Current research focuses on understanding the association between these pathologies and exploring potential therapeutic options.

Mouse periodontitis models using whole Porphyromonas gingivalis bacteria induction

Ninuk Hariyani et al.

Summary: This study presents a scoping review of the current literature regarding the available method to produce mouse periodontitis models using whole Porphyromonas gingivalis bacteria. The most commonly used vehicle to induce a mouse periodontitis model is the combination of P. gingivalis with ligature. Future research on different types of vehicles and bacteria for inducing more effective and more time-efficient periodontitis models is needed.

GPR40 deficiency is associated with hepatic FAT/CD36 upregulation, steatosis, inflammation, and cell injury in C57BL/6 mice

Zhongyang Lu et al.

Summary: Deficiency of GPR40 does not alleviate high-fat diet-induced NAFLD, but instead increases metabolic abnormalities and hepatic steatosis under a low-fat diet. GPR40 deficiency leads to upregulation of hepatic fatty acid translocase (FAT)/CD36 expression, possibly inhibiting the development of nonalcoholic steatohepatitis by modulating FAT/CD36 expression.

Targeting lipid GPCRs to treat type 2 diabetes mellitus - progress and challenges

Julien Ghislain et al.

Summary: This Review summarizes the physiological functions, pharmacology, and clinical studies of G protein-coupled receptors specific for lipid metabolite ligands and discusses the challenges associated with developing therapeutics for type 2 diabetes mellitus. Currently, therapeutic approaches targeting these receptors focus on increasing insulin secretion either by directly acting on beta-cells or indirectly on gastrointestinal enteroendocrine cells that release hormones regulating insulin secretion.

Intersection between macrophages and periodontal pathogens in periodontitis

Wenzhe Wang et al.

Summary: The interaction between macrophages and periodontal pathogens plays a crucial role in the development of periodontal disease. Macrophages are key players in maintaining tissue homeostasis and immune response. Macrophages may influence the extent of tissue destruction in periodontal tissues through pro-inflammation and anti-inflammation phenotypes.

Association between metabolic syndrome and periodontitis: The role of lipids, inflammatory cytokines, altered host response, and the microbiome

Flavia Q. Pirih et al.

Summary: Periodontitis is associated with metabolic syndrome, diabetes, and cardiovascular disease, with evidence suggesting that these conditions can alter the oral microbiome. Studies indicate an interrelationship between periodontitis and metabolic syndrome, where periodontitis may contribute to or worsen metabolic syndrome. Additional research is needed to fully comprehend the impact of these conditions on each other.
