Biomimetic Peptide Catalytic Bond-Forming Utilizing a Mild Bronsted Acid

Synthesis of tryptophan-dehydrobutyrine diketopiperazine and biological activity of hangtaimycin and its co-metabolites

Summary: An improved synthesis method for tryptophan-dehydrobutyrine diketopiperazine (TDD), a co-metabolite of the polyketide/non-ribosomal peptide hybrid antibiotic hangtaimycin starting from γ-tryptophan, is presented. Comparison with TDD isolated from Streptomyces spectabilis, the producer of hangtaimycin, confirmed its S configuration. The X-ray structure analysis of the racemate revealed an interesting dimerization through hydrogen bonds. The bioactivity of hangtaimycin, TDD, and the hangtaimycin degradation product HTM222 was also tested.

Epipolythiodioxopiperazine-Based Natural Products: Building Blocks, Biosynthesis and Biological Activities

Recent studies have elucidated the principles of ETP production, including sulfurization via glutathione, disulfide bond cleavage, and subsequent oxidation or methylation reactions, which impact the expression of biosynthesis clusters, metabolite toxicity and secretion, and self-tolerance of the producer.

Organocatalytic Activation of Inert Hydrosilane for Peptide Bond Formation

Summary: We describe the development of a reliable catalytic protocol for peptide bond formation that is generally applicable to natural and unnatural alpha-amino acids, beta-amino acids, and peptides bearing various functional groups. A 10 mol % loading of HSi[OCH(CF3)(2)](3) as a catalyst was sufficient to guarantee a consistently high yield of the resulting peptide. This method facilitates the sustainable utilization of natural resources by using a catalyst and an auxiliary based on earth-abundant silicon.
