A TNF receptor 2 agonist ameliorates neuropathology and improves cognition in an Alzheimer's disease mouse model

Summary: This study investigated the effects of stimulating TNFR2 with a TNFR2 agonist (NewStar2) in an AD mouse model. The results demonstrated that activation of TNFR2 reduced amyloid beta deposition and expression of beta-secretase 1, increased microglial and astrocytic activation, promoted the uptake and degradation of A beta, and improved cognitive functions. These findings suggest that TNFR2 stimulation might be a potential treatment for AD.

Summary: Neuronal TNF receptors play distinct roles in CNS pathology in vivo, with TNFR1 and IKK beta promoting microglial inflammation and neurotoxicity in demyelination, while TNFR2 mediates neuroprotection in excitotoxicity. The neuroprotective effects of solTNF against glutamate excitotoxicity in vitro are indirect and mediated via astrocyte tmTNF-neuron TNFR2 interactions. Selective inhibition of solTNF/TNFR1 with maintenance of TNFR2 function could have combined anti-inflammatory and neuroprotective properties for safe treatment of CNS diseases.

Future TNF-targeting drugs may focus on selectively blocking TNF binding to TNFR1 and on TNFR2 agonists to improve treatment outcomes for rheumatic diseases.
