TNF signaling pathway-mediated microglial activation in the PFC underlies acute paradoxical sleep deprivation-induced anxiety-like behaviors in mice

Haiying Liu et al.

Summary: The study shows significant correlations between anxiety-like behavior, microglial activation, and inflammation during acute sleep deprivation. The research confirmed alterations in key genes related to the TNF signaling pathway and demonstrated changes in microglial cells in the prefrontal cortex in a mouse model of sleep deprivation. Additionally, the study suggests that minocycline, an anti-inflammatory agent, may alleviate the anxiogenic effect and microglial activation.

Fluorocitrate and neurotropin confer analgesic effects on neuropathic pain in diabetic rats via inhibition of astrocyte activation in the periaqueductal gray

Xingfeng Liu et al.

Summary: Effective treatments for diabetic neuropathic pain (DNP) are still unmet clinical needs. Activating astrocytes in the ventrolateral region of periaqueductal gray (vlPAG) can regulate pain responses, and inhibiting astrocyte activation in vlPAG produces an analgesic effect on DNP. In a rat model, injecting fluorocitrate (FC) or neurotropin in vlPAG attenuated DNP symptoms by suppressing astrocyte activation.

Microglia in depression: current perspectives

Xiaoning Jia et al.

Summary: This review focuses on the role of microglia in the etiology of depression, discussing the dynamic states of microglia, the correlative and causal evidence of microglial abnormalities in depression, possible mechanisms of how microglia sense depression-related stress and modulate depression state, and how antidepressive therapies affect microglia. Understanding the role of microglia in depression may lead to new treatment strategies for fighting against this mental illness.

On inflammatory hypothesis of depression: what is the role of IL-6 in the middle of the chaos?

Elnaz Roohi et al.

Summary: Alterations in IL-6 levels may contribute to depression symptomatology in various ways, both in the periphery and the brain. IL-6 acts on multiple target tissues throughout the body, and dysregulation of this cytokine can lead to events relevant to depression. Blocking the effects of IL-6 may prevent further escalation of inflammatory responses and open new avenues in diagnosis, treatment, and prevention of this debilitating disorder.

Macrophages inhibit and enhance endometriosis depending on their origin

Chloe Hogg et al.

Summary: Macrophages play an important role in the pathophysiology of endometriosis, with lesion-resident macrophages derived from eutopic endometrial tissue, infiltrating large peritoneal macrophages, and monocytes. Continuous recruitment of monocytes and expansion of CCR2+ large peritoneal macrophages are triggered by endometriosis. Depletion of eutopic endometrial macrophages results in smaller lesions, while inhibiting monocyte recruitment reduces macrophage populations and increases lesion number. Reprogramming the ontogeny of peritoneal macrophages decreases lesion development. The study suggests a model where endometrial macrophages promote endometriosis while newly recruited monocyte-derived macrophages, possibly in large peritoneal macrophage form, have antiendometriotic effects.

A mouse model of endometriosis that displays vaginal, colon, cutaneous, and bladder sensory comorbidities

Joel Castro et al.

Summary: This study utilized an autologous mouse model of endometriosis to examine disease features and symptoms including lesion development, chronic inflammatory environment, neuroangiogenesis, sensory hypersensitivity, and altered pain responses. The study found significant increases in lesion size, endometrial glands, stroma, and neuroangiogenesis. Mice with endometriosis displayed enhanced pain sensitivity and altered bladder function compared to control mice.

Implication of cerebral astrocytes in major depression: A review of fine neuroanatomical evidence in humans

Liam Anuj O'Leary et al.

Summary: Postmortem investigations have revealed lower expression levels and densities of astrocytes in individuals with major depressive disorder (MDD), suggesting a loss of astrocytes in this mental illness. Studies comparing astrocyte density and morphometry show that human subcortical brain regions have more abundant and smaller astrocytes than cortical regions. These findings provide insight into the potential role of astrocyte pathology in MDD and suggest a hypothesis for the antidepressant effects of ketamine.

Sex-dependent Cav2.3 channel contribution to the secondary hyperalgesia in a mice model of central sensitization

Marcella Amorim Ferreira et al.

Summary: Central sensitization, a characteristic of difficult to treat painful conditions, involves calcium influx in nociceptive neurons, with the Cav2.1, Cav2.2, and now the Cav2.3 voltage-gated calcium channels playing key roles. Inhibition of Cav2.3 has shown to partially reverse secondary hyperalgesia in a mouse model, especially in female mice, indicating its potential as a molecular target for CS. Additionally, the role of sexual differences in painful conditions needs more attention and exploration in clinically relevant pain models.

Summary: Endometriosis is a gynecological disease that affects women of reproductive age, causing gynecologic symptoms and systemic effects like inflammation, altered body weight, and behavioral changes. Research indicates that endometriosis may alter the activity level of the hypocretin/orexin neuronal system, leading to low body weight, differences in body composition, and behavioral changes.

Reactive Astrocytes: Critical Players in the Development of Chronic Pain

James Tang et al.

Summary: Chronic pain is associated with long term plasticity of nociceptive pathways in the central nervous system. Reactive astrogliosis in astrocytes alters their release of cytokines and gliotransmitters, potentially driving persistent changes in central nociceptive activity. Lactate released by astrocytes may play a role in modulating neuronal and astrocytic activity, supporting pathological changes in central nociceptive activity.
