Chronic alcohol metabolism results in DNA repair infidelity and cell cycle-induced senescence in neurons

Summary: Chronic binge-like drinking is a risk factor for age-related dementia, potentially due to its lasting and irreversible impact on the brain. Ethanol exposure leads to pro-ageing changes in brain cortices, particularly in neurons, by affecting the ethyl alcohol oxidation and folate metabolism. This disrupts DNA repair pathways, forces post-mitotic cells to re-enter a cell cycle-like process, and ultimately results in irreversible senescence response. Supplementation of bioactive 5-methyl tetrahydrofolate during alcohol exposure can prevent these effects, highlighting the connection between binge drinking and dementia.
