Observation of conformational changes that underlie the catalytic cycle of Xrn2

Summary: In this study, we used NMR methods to investigate the structure and dynamics of eukaryotic 5'->3' exoribonuclease Xrn2. The results showed that the enzyme exhibited high dynamics around the catalytic center and that the conformational equilibrium shifted towards the active state in the presence of substrate and magnesium. Additionally, the dynamics in Xrn2 were found to correlate with the RNA degradation rate.

Research on ARNT PAS-B Y456T demonstrates that the interconversion process supports two key hypotheses: the slipped conformation is smaller and less compressible, and the process proceeds through a chiefly unfolded intermediate state.

Summary: Imidazole glycerol phosphate synthase (HisFH) is a heterodimeric bienzyme complex responsible for catalyzing the hydrolysis of glutamine and cyclase reaction. The catalytically active HisFH conformation is formed only when both substrates of HisH and HisF are bound, leading to allosteric activation. In solution, inactive and active conformations are dynamically balanced, with HisFH turnover rates correlating with the active conformation population.
