Structure-Aided Design, Synthesis, and Biological Evaluation of Potent and Selective Non-Nucleoside Inhibitors Targeting Protein Arginine Methyltransferase 5

Summary: In this study, a novel series of non-nucleoside PRMT5 inhibitors were designed through structure-aided drug design approach. Compound 41 was discovered as a highly potent and selective PRMT5 inhibitor, which exerted anti-proliferative effects and inhibited the methyltransferase activity of PRMT5 in cells. Additionally, compound 41 showed attractive pharmacokinetic properties and effectively suppressed tumor growth in an animal model.
